HCT116 and LoVo cells were cultured in DMEM supplemented with 10% FBS and 1% antibiotics at 37°C with 5% CO₂. Transcription profiles and clinical info for colorectal adenocarcinoma (COAD) were downloaded from TCGA and GEO databases. SiRNAs and plasmids were transfected using Lipofectamine 2000. Single-cell transcriptome profiles were analyzed using Seurat, and gene set enrichment analysis (GSEA) was performed with GSEA software. WGCNA was used to determine gene co-expression modules. Western blotting, qRT-PCR, Transwell assay, ChIP-seq, and ChIP-qPCR were conducted to analyze protein and gene expression. FISH and immunofluorescence were used to detect LINC00887 RNA probe. Immunohistochemistry (IHC) was performed on CRC tissue chips. Animal experiments were approved by IACUC, and statistical analysis was done using GraphPad Prism software.
