RIG-101 is an intranasal stem-loop RNA (SLR) therapeutic that agonizes RIG-I, triggering type I and type III interferon production in the respiratory epithelium. It is being developed by Leyden Labs, which acquired its originator RIGImmune in spring 2026, initially to prevent respiratory viruses from triggering asthma exacerbations. A completed Phase I study showed acceptable safety and on-target pharmacodynamic activity in healthy volunteers and asthma patients, and a two-part Phase I/II human rhinovirus RV-A16 challenge trial in adults with asthma is recruiting.

RIG-101 is an oligonucleotide of the stem-loop RNA therapeutic (SLR) class that agonizes RIG-I, a cytosolic innate immune receptor. Activation of RIG-I triggers type I and type III interferon production in the respiratory epithelium, priming innate antiviral defenses at the mucosal surface. In asthma patients, intranasal RIG-101 induced type I and type III interferons and upregulated interferon-stimulated genes in the upper respiratory tract, a pharmacodynamic signature consistent with the intended mechanism of action.
