Trastuzumab botidotin binds HER2 on the surface of tumor cells and is internalized; the stable enzyme-cleavable linker releases the MMAF-derivative payload Duo-5 (Duostatin-5), a tubulin inhibitor that induces G2/M cell-cycle arrest and apoptosis. The trastuzumab moiety also inhibits HER2 signaling and mediates antibody-dependent cell-mediated cytotoxicity (ADCC).
Unresectable or metastatic HER2-positive breast cancer in adult patients who have received one or more prior anti-HER2 therapy (approved by China's NMPA in October 2025).
