Novel Mechanism of Action
Procizumab is designed to selectively target circulating dipeptidyl peptidase 3 (搜索) (cDPP3 (搜索)), a cardiac depression factor and key pathological driver in shock. Under normal physiological conditions, DPP3 functions as an intracellular enzyme. However, when released into circulation following cellular injury, it degrades angiotensin peptides, resulting in dysregulation of the renin-angiotensin-aldosterone system (搜索) (RAAS). This loss of RAAS control can lead to shock, broad organ failure, and ultimately death.
By inhibiting cDPP3 (搜索) activity, procizumab restores RAAS balance and stabilizes cardiovascular function. The therapeutic potential has been demonstrated in preclinical and clinical settings, where it effectively normalized cardiovascular parameters, reversed organ dysfunction, and increased survival while exhibiting a favorable safety and tolerability profile in a completed Phase 1 study in healthy volunteers.
Clinical Development Progress
The ongoing multicenter, randomized, double-blind, placebo-controlled Phase 2a PROCARD 2a trial is evaluating the safety, tolerability, pharmacokinetics, and pharmacodynamics of procizumab in patients with cardiogenic shock (搜索) and elevated cDPP3 (搜索) levels. The additional funding will support continued enrollment and expand the study footprint across Europe while preparing for planned expansion into the United States.
