The company projects approximately $1.7 billion in global net sales by 2028, driven by its commercial portfolio anchored by NUPLAZID and DAYBUE.
Strong Commercial Growth Trajectory
The projected 2028 revenue breakdown includes approximately $1 billion from NUPLAZID (pimavanserin) for Parkinson's disease psychosis (搜索) and ~$700 million from DAYBUE (trofinetide) for Rett syndrome (搜索). This represents substantial growth for the company's commercial engine, which currently includes the first and only FDA-approved treatments for both indications.

Alzheimer's Disease Psychosis Program Advances
The company's most significant near-term catalyst involves remlifanserin (搜索) (formerly ACP-204), with top-line results from the Phase 2 RADIANT study in Alzheimer's disease psychosis (搜索) expected between August and October 2026. This program represents a substantial commercial opportunity, with remlifanserin expected to account for approximately $4 billion of the $11 billion unadjusted full peak sales opportunity represented by Acadia's current pipeline.
The $4 billion projection encompasses remlifanserin (搜索)'s potential in both Alzheimer's disease psychosis (搜索) and Lewy body dementia psychosis (搜索), addressing significant unmet medical needs in neuropsychiatric disorders.
DAYBUE STIX Launch and Global Expansion
Acadia received FDA approval on December 12, 2025, for DAYBUE STIX (搜索) (trofinetide) for oral solution, a dye- and preservative-free powder formulation for treating Rett syndrome (搜索) in adult and pediatric patients 2 years and older. The product will be available on a limited basis starting in Q1 2026, with a broader launch planned for Q2 2026.
"The introduction of DAYBUE STIX (搜索) in the U.S. represents an exciting opportunity to deliver a more flexible option for patients and caregivers, further strengthening our leadership position in Rett syndrome (搜索) care," Adams noted.
The company is also expanding DAYBUE's global reach, with Israel's Ministry of Health recently approving DAYBUE oral solution for Rett syndrome (搜索) treatment in adults and pediatric patients 2 years of age and older weighing at least 9 kg. An opinion from the European Medicines Agency's Committee for Medicinal Products for Human Use (CHMP) for trofinetide is expected in Q1 2026.
Robust Clinical Pipeline Development
Acadia's R&D pipeline continues advancing with plans to initiate five Phase 2 or Phase 3 studies across eight disclosed programs and deliver four Phase 2 or Phase 3 readouts by the end of 2027. Top-line results from a Phase 3 study in Japan of trofinetide for Rett syndrome (搜索) treatment are anticipated between October 2026 and March 2027.
Mechanism of Action and Therapeutic Approach
NUPLAZID (pimavanserin) functions as a selective serotonin inverse agonist and antagonist preferentially targeting 5-HT2A receptors (搜索), which are thought to play an important role in neuropsychiatric disorders. The drug demonstrates no appreciable binding affinity for dopamine, histamine, muscarinic, or adrenergic receptors, providing a targeted approach to treating hallucinations and delusions associated with Parkinson's disease psychosis (搜索).
DAYBUE (trofinetide) represents a synthetic version of the naturally occurring tripeptide glycine-proline-glutamate (GPE), though the mechanism by which trofinetide exerts therapeutic effects in Rett syndrome (搜索) patients remains unknown.
