The partnership aims to reduce supply chain risks and manufacturing costs while establishing domestic production capacity ahead of the anticipated commercial launch in the first half of 2027.
Manufacturing Partnership Addresses Supply Chain Concerns
The collaboration with Adare Pharma Solutions (search) comes as Achieve seeks to mitigate risks associated with international pharmaceutical importation and potential tariffs. The company has already commenced technology transfer to Adare, which will provide supply chain redundancy and U.S.-based contingency manufacturing capacity.

The partnership decision was influenced by recent FDA inspection findings at one of the manufacturers named in cytisinicline's New Drug Application (NDA). The FDA's current Good Manufacturing Practices inspection identified two observations related to solid oral dose manufacturing, which are being addressed through ongoing communications with the agency regarding remedial action plans.
Regulatory Progress and Priority Status
The FDA accepted cytisinicline's NDA for treatment of nicotine dependence (search) for smoking cessation in adults, assigning a Prescription Drug User Fee Act (PDUFA) targeted action date of June 20, 2026. The application is based on two successfully completed Phase 3 studies and an open-label safety study.
Significantly, cytisinicline was selected as one of the first nine therapies chosen for the inaugural FDA Commissioner's National Priority Voucher for e-cigarette or vaping cessation. This designation provides enhanced FDA communications and expedited review once complete materials are submitted to the agency.
The company completed the ORCA-OL long-term exposure trial with 334 participants finishing the one-year study, providing comprehensive long-term safety data that significantly exceeds the number of patients required for FDA review.
Clinical Evidence Supports Broad Efficacy
Recent publications have strengthened the clinical evidence supporting cytisinicline's effectiveness across diverse patient populations. Complete results from the ORCA-3 trial, published in the Journal of the American Medical Association (JAMA) Internal Medicine, reaffirmed cytisinicline's efficacy and tolerability for smoking cessation in adult smokers at both 6- and 12-week treatment durations, including reduction in nicotine cravings and extended cessation benefits through 24 weeks.
Additional data published in Thorax demonstrated that cytisinicline significantly increased continuous smoking abstinence versus placebo in both chronic obstructive pulmonary disease (search) (COPD) and non-COPD subgroups. Despite more severe tobacco use histories and greater prior treatment exposure, participants with COPD achieved quit rates comparable to those without COPD.
"The SRNT findings are particularly encouraging because they indicate that cytisinicline helps people quit smoking regardless of their prior treatment history or number of prior attempts, and, if approved, could offer hope to those who have struggled to quit," said Mark Rubinstein, MD, Chief Medical Officer of Achieve.
New data presented at the Society for Research on Nicotine and Tobacco (SRNT) 2026 Annual Meeting indicated that cytisinicline delivers meaningful quitting success regardless of participants' prior use of smoking cessation medications or number of previous quit attempts.
Addressing Significant Public Health Need
Cytisinicline targets a substantial unmet medical need, with approximately 25 million adults in the United States smoking combustible cigarettes and nearly 18 million adults using e-cigarettes. Tobacco use remains the leading cause of preventable death, responsible for more than eight million deaths worldwide and nearly half a million deaths in the United States annually.
More than 87% of lung cancer (search) deaths, 61% of all pulmonary disease deaths, and 32% of all deaths from coronary heart disease (search) are attributable to smoking and exposure to secondhand smoke. Currently, there are no FDA-approved treatments indicated specifically as an aid to nicotine e-cigarette cessation.
Mechanism of Action and Commercial Preparation
Cytisinicline is a plant-based alkaloid with high binding affinity to the nicotinic acetylcholine receptor (search). It is believed to aid in treating nicotine addiction by interacting with nicotine receptors in the brain, reducing the severity of nicotine craving symptoms, and reducing the reward and satisfaction associated with nicotine products.
The company has completed supply chain and market access readiness activities, including implementation of third-party logistics providers and specialty pharmacy partner selection. Achieve has also announced a partnership with Omnicom (search) across multiple agencies to create a data-driven commercial model designed to execute with precision and scale efficiently.
Financial Position
As of December 31, 2025, Achieve reported cash, cash equivalents, and marketable securities of $36.4 million. Total operating expenses for the twelve months ended December 31, 2025, were $54.9 million, with a total net loss of $54.7 million for the same period.
