The oversubscribed financing round was co-led by RA Capital Management (search) and Patient Square Capital (search)'s platform Enavate Sciences (search), with participation from Abiogen Pharma (search), Janus Henderson Investors, Arkin Bio, Balyasny Asset Management, Transhuman Capital, Adage Capital Partners LP, and other dedicated life sciences investors.
FDA Designations Support Accelerated Development Path
The U.S. Food and Drug Administration has granted neridronate breakthrough therapy, fast track, and orphan drug designations for the treatment of CRPS. These regulatory designations are expected to facilitate expedited development and review processes for the therapy, which targets a rare disease with significant unmet medical need.
CRPS-1 (search) is a severe, debilitating rare disease with an estimated 65,000 new cases annually in the United States. The condition is characterized by intense, continuous pain in the affected limb, typically in extremities such as the arm, leg, hand or foot. Patients experience an evolving condition that begins with a "warm" phase lasting approximately six to twelve months after initial injury, where inflammation causes the affected limb to become red, swollen, warm, and painful, progressing to a chronic "cold" phase with ongoing, debilitating pain.
Proven Track Record in Italian Market
Ambros licensed the rights to neridronate from Italian pharmaceutical company Abiogen Pharma (search) S.p.A. under a strategic collaboration, providing exclusive rights to neridronate in North America with an option for broader market expansion. Neridronate was approved in Italy based on safety and efficacy demonstrated in two CRPS Phase 3 studies and has been used for CRPS and other disorders in more than 600,000 patients to date.

Novel Bisphosphonate Mechanism
Neridronate is a novel bisphosphonate (search) that was discovered, developed, and commercialized by Abiogen Pharma (search) S.p.A. for the Italian market. The drug belongs to a class targeting bisphosphonate, which is believed to have a role in preventing soft tissue calcification and in regulating bone mineralization. Clinical studies have demonstrated lasting pain reduction along with improvements in other CRPS-related symptoms.
Beyond CRPS, neridronate is approved in Italy for additional indications including osteogenesis imperfecta (search) and Paget's disease (search). Its well-established safety profile and therapeutic benefits position it as a potential promising treatment for patients with CRPS-1 (search) worldwide.
Phase 3 Trial Timeline and Leadership
The financing is expected to support the neridronate pivotal Phase 3 clinical trial in CRPS-1 (search) (CRPS-RISE) and related regulatory preparations and pre-commercial activities. The pivotal Phase 3 trial is planned to start enrollment in the first quarter of 2026 and represents a potential first-in-class approach to addressing a disease with no FDA-approved medicines.
