The research, led by teams at the Royal Melbourne Institute of Technology (RMIT) and the Centre for Eye Research Australia (CERA), demonstrates a novel nano-delivery system that successfully transports therapeutic compounds to the back of the eye.
Revolutionary Nano-Cubosome Technology
The innovation centers on cubosomes, nanoparticles with a unique internal structure capable of carrying both water-loving and fat-loving molecules. Researchers loaded these nano-carriers with lutein, a natural antioxidant extracted from Gac fruit that concentrates in the macula and helps filter high-energy blue light while reducing oxidative stress.
"Our cubosome carriers act like tiny shields, keeping the compound safe and releasing it in a controlled way once it's inside the eye," said co-lead researcher Charlotte Conn from RMIT's School of Science.
The team overcame a major challenge in ocular drug delivery by blending lutein with a cationic lipid called MO-DOTAP (search), creating particles with the optimal size, surface charge, and adhesive properties to penetrate the eye's natural barriers and remain long enough for therapeutic effect.
Promising Preclinical Results
In studies using Swiss ARC mice with naturally low retinal lutein levels, researchers compared intravitreal injections with the experimental eyedrops. While injections achieved peak concentrations within one hour, the eyedrops demonstrated steady lutein accumulation in retinal tissue over one week, suggesting potential for long-term, needle-free delivery.
Laboratory testing on retinal pigment epithelial cells revealed significant therapeutic benefits. When these cells were subjected to oxidative stress, they typically produce high concentrations of toxic compounds VEGF-A (search) and IL-6, which promote inflammation and pathological blood vessel development. Treatment with lutein cubosomes reduced concentrations of both compounds through activation of the Nrf2/HO-1 (search) pathway, a natural antioxidant defense mechanism.
Clinical Implications and Patient Benefits
"Frequent eye injections are uncomfortable and can be distressing for patients," explained Dr. Dao Nguyen, who co-led the research at RMIT and is now at Deakin University's School of Medicine. "People could use the eyedrop as a preventative measure that could reduce the risk of developing late-stage diseases and the need for injections."
Current treatment for retinal diseases (search) involves repeated injections directly into the eye, a procedure that is painful, costly, and stressful for patients. The new eyedrop formulation remained stable at room temperature for months, making it practical for real-world use and potentially improving patient compliance.
Broad Therapeutic Platform
Beyond lutein delivery, the cubosome technology represents a versatile platform for ocular drug delivery. "This is a technology with broad potential," said Associate Professor Tien Huynh from RMIT's School of Science. "We've shown it can protect fragile ingredients and carry them safely to the back of the eye, which has long been a barrier for treatments."
The delivery system could potentially transport various sensitive medications to treat different retinal conditions, opening new therapeutic possibilities for diseases that currently require invasive procedures.
Current Limitations and Future Directions
The research remains in early stages, with studies conducted only on healthy mice and cell cultures rather than disease models. Some limitations emerged during testing, including instances where eyedrops failed to remain on the animal's eye or were rubbed off, reducing effectiveness. Additionally, the thinner outer eye layer in mice compared to humans may affect the transferability of results.
"This kind of approach could transform how we manage age-related macular degeneration (search)," said Associate Professor Chi Luu from CERA and the University of Melbourne. "If future trials confirm the safety and effectiveness of the delivery platform, eyedrops could one day be used for treating early stages of AMD and other serious retinal diseases (search)."
The research team plans to conduct testing in larger animal models before collaborating with industry and clinical partners to develop human trials. While the eyedrops may not completely replace injections, successful development could delay or prevent the need for more invasive treatments in retinal disease management.
The findings were published in ACS Applied Materials & Interfaces, marking a significant step toward more patient-friendly treatments for sight-threatening retinal conditions.
