Currently, 9 out of 10 young patients who experience relapse succumb to the disease. In high-risk cases, 15% of children do not respond to initial treatment, and half of those who do respond initially will see their cancer return.
"The statistics once patients get to that point are devastating for families," said Associate Professor David Croucher from the Garvan Institute. "Behind every statistic is someone's loved one."
Overcoming Cellular Resistance Mechanisms
The research team discovered that standard chemotherapy (search) drugs rely on a specific cellular pathway called the JNK pathway (search) to trigger cancer cell death. In relapsed tumors, this critical "switch" often stops functioning, rendering conventional treatments ineffective and allowing tumors to spread with deadly consequences.
"By finding drugs that don't depend on the JNK pathway (search), we can still trigger cancer cell death even when this usual route is blocked," Croucher explained.
Promising Preclinical Results
The findings, published in Science Advances, demonstrate that romidepsin combined with standard chemotherapy (search) can halt tumor growth through alternative cell-death pathways. In animal model studies, this combination approach showed multiple therapeutic benefits:
