Among patients with BSA ≥10%, the EASI-75 response rates were 61.1% for the 0.3% group, 46.2% for the 1.0% group, and 20% for the vehicle group.
The vIGA-AD response rates (score 0/1 with ≥2-point improvement) further supported the drug's efficacy, with rates of 44.4%, 38.5%, and 10% for the 0.3%, 1.0%, and vehicle groups respectively in the same patient population. Notably, patients with BSA ≥20% demonstrated even greater clinical improvements with both LNK01004 groups, reinforcing the drug's potential for treating more severe disease presentations.
Safety Profile Demonstrates Minimal Systemic Exposure
LNK01004 exhibited excellent tolerability with remarkably low systemic exposure following topical administration. Mean maximum plasma concentrations (Cmax) were 0.06 ng/mL and 0.15 ng/mL for the 0.3% and 1.0% groups respectively, representing levels hundreds of times lower than the human whole-blood IC50.
All treatment-related adverse events were classified as mild or moderate (Grade 1-2), and no treatment-related serious adverse events were observed throughout the study period. The safety profile remained consistent with findings from earlier clinical studies.
