Boehringer Ingelheim Partners with EMFRET Analytics to Advance Novel GPVI Inhibitor EMA601 for Stroke Treatment

Novel Mechanism Targets Platelet Dysfunction in Stroke
EMA601 specifically targets glycoprotein VI (search) (GPVI (search)), a platelet surface receptor that plays a central role in thrombosis (search) and infarction but is not required for normal blood coagulation. The receptor, found exclusively on thrombocytes and their precursor cells in bone marrow, binds to exposed collagen on injured vessel walls, triggering platelet activation and aggregation.
"Our GPVI (search) inhibitor EMA601, discovered in Würzburg, shows significant efficacy," says Professor Bernhard Nieswandt from University Hospital Würzburg (search). "EMA601 is a very potent GPVI inhibitor and has therefore the potential to offer clinical advantages."
The scientific foundation for EMA601 dates back to 2001, when Nieswandt and his doctoral student Valerie Orth first described GPVI (search)'s central role while working at Witten/Herdecke University. Their groundbreaking research demonstrated that neutralizing GPVI in mouse models using monoclonal antibodies resulted in protection against thrombosis (search) without significantly disturbing normal hemostasis.
Preclinical Evidence Demonstrates Therapeutic Promise
In November 2024, researchers published compelling preclinical data in the European Heart Journal demonstrating EMA601's therapeutic potential. The study, conducted through a combination of biochemical tests, cell-based assays, and animal models, showed three key findings: EMA601 selectively inhibits GPVI (search) signaling without shutting down the coagulation system, GPVI blockade prevented pathological clot formation in mouse models, and the treatment reduced both thrombosis (search) and inflammation-driven tissue damage following ischemia.
A 2007 study had already established the therapeutic foundation for GPVI (search) blockade in stroke (search) treatment. Nieswandt, working with Guido Stoll and other researchers, discovered that GPVI blockade significantly reduced infarct size in the brain, improved neurological outcomes, and crucially, did not increase the risk of intracranial bleeding in preclinical stroke models.
Addressing Thrombo-inflammation in Stroke Treatment
The collaboration addresses a critical gap in current stroke (search) therapy. Despite significant advances in treatment—from intravenous thrombolysis introduced in Europe in 1995 to endovascular clot removal—stroke therapy remains limited. In approximately half of successfully recanalized patients, restoring blood flow alone is insufficient to achieve good functional outcomes.
"In animal studies, we were able to show that an inflammatory process—thrombo-inflammation—is triggered in the downstream hypoperfused brain areas as soon as vessel occlusion begins. This process remains active despite recanalization and allows infarcts to continue growing," explains Professor Guido Stoll.
EMA601 may serve as an adjunct therapy to thrombolysis or thrombectomy, as it demonstrated the ability to inhibit further infarct growth after recanalization in humanized GPVI (search) mouse models. A major advantage is the low bleeding risk shown in preclinical studies, as EMA601 does not appear to impair normal blood coagulation—a critical consideration in acute stroke (search) where life-threatening intracranial hemorrhages can occur.
Strategic Partnership Aims for First-in-Class Development
The cooperation positions EMA601 as a potential first-in-class compound that could transform antithrombotic and anti-inflammatory therapy. "Boehringer Ingelheim has shaped acute stroke (search) care for decades," says Søren Tullin, Senior Vice President and Global Head of Cardiometabolic Diseases Research at Boehringer Ingelheim. EMFRET Analytics (search), founded in 2002 as a bootstrapped company, developed its program independently without external capital. "We developed, produced, and distributed antibodies and reagents for research worldwide," reports Dr. Valerie Orth, CEO of EMFRET Analytics. Addressing Global Stroke Burden
The partnership addresses a significant global health challenge. Stroke (search) remains one of the most common causes of death and disability, with approximately 11.9 million new cases occurring annually and 93.8 million people living with long-term consequences. Due to aging and population growth, the global burden of stroke is expected to continue rising.
