Bone Biologics Doubles Australian Trial Sites for NB1 Spinal Fusion Study, Targets Year-End 2026 Enrollment
核心洞察
Bone Biologics has expanded its first-in-human pilot trial of NB1 to six Australian hospital sites, doubling the number of participating centers.
The multicenter, randomized study evaluates NB1 in up to 30 adults with degenerative disc disease (搜索) undergoing transforaminal lumbar interbody fusion.
The company expects to complete patient enrollment by year-end 2026, with fusion success and Oswestry Disability Index change as primary endpoints.
Bone Biologics Corporation (Nasdaq: BBLG, BBLGW) announced that six hospital sites in Australia are now under agreement to participate in the first-in-human pilot clinical trial of NB1, its rhNELL-1 (搜索)-based bone graft product candidate for spinal fusion. The expanded network doubles the number of participating centers and is expected to accelerate patient screening and surgical scheduling, strengthening the company's expectation to complete enrollment by year-end 2026.
NB1 combines the recombinant human NELL-1 (搜索) protein (rhNELL-1 (搜索)) with demineralized bone matrix for use in spinal procedures. The Burlington, Massachusetts-based company is developing orthobiologic products for spine fusion markets.
"We believe the additional hospital sites in our study meaningfully increases our access to qualified patients and reduces our reliance on patient flow at any individual center," said Bone Biologics CEO Jeff Frelick. "This expansion strengthens our anticipation of reaching full enrollment by the end 2026 and brings us closer to the 12-month follow-up data."
Trial Design and Endpoints
The pilot study is a multicenter, prospective, parallel-group, randomized trial conducted in Australia that evaluates the safety and preliminary effectiveness of NB1 in up to 30 adult subjects with degenerative disc disease (搜索) (DDD) undergoing transforaminal lumbar interbody fusion (TLIF).
Inclusion criteria include patients with DDD at one level from L2-S1 who may also have up to Grade 1 spondylolisthesis (搜索) or Grade 1 retrolisthesis (搜索) at the involved level. Primary clinical endpoints include fusion success at 12 and 24 months following surgery, and change from baseline in the Oswestry Disability Index, a standard measure of functional disability.
The U.S. Food and Drug Administration (搜索)'s Division of Orthopedic Devices previously reviewed the study design in a pre-submission, and these pilot study findings are intended to support progression to a pivotal clinical study in the United States.
Manufacturing and Stability Milestones
The company has reported sequential extensions of the validated shelf life of its rhNELL-1 (搜索) protein. In May 2026, Bone Biologics announced that the validated shelf life had been extended to 29 months, building on the 24-month milestone achieved in December 2025. In its September update, the company referenced a 32-month validated shelf life for rhNELL-1.
"Supported by our cash runway through the second quarter of 2027 and our recently announced 32-month validated shelf life for rhNELL-1 (搜索), we remain focused on disciplined execution of the NB1 clinical program as we prepare for a potential pivotal study," Frelick added.
Capital Position and 2026 Priorities
In July 2026, the company completed a private placement with a single healthcare-focused institutional investor, priced at-the-market under Nasdaq rules, generating gross proceeds of approximately $3.0 million before deducting placement agent fees and offering expenses. The financing also included Series F warrants and Series G warrants which, if fully exercised on a cash basis, would provide up to approximately $6.0 million of additional gross proceeds. The company noted that no assurance can be given that any of the warrants will be exercised or that it will receive cash proceeds from their exercise.
Net proceeds are intended to fund clinical trials, maintain and extend the patent portfolio, and support working capital and general corporate purposes. Bone Biologics believes the financing extends its operating runway through Q2 2027, providing the capital base to execute clinical and operational priorities without near-term reliance on additional financing.
In January 2026, the company outlined its priorities for the year: advancing the NB1 clinical program, continuing manufacturing and regulatory readiness efforts, pursuing additional intellectual property protection for its rhNELL-1 (搜索) technology, and maintaining capital discipline. The mid-year update stated that progress through the first half of 2026 reflected execution against each of these priorities.
"Spine fusion remains an underserved market and closing that gap with a differentiated biologic is the opportunity in front of us," Frelick said. "Our shareholders' support has been foundational to the progress we've made getting here. The mandate for the remainder of 2026 is unchanged: execute the clinical program with discipline and manage our capital with the same rigor."
Development Scope
Bone Biologics was founded to pursue regenerative medicine for bone. The company is undertaking the clinical study in Australia with select strategic partners, building on preclinical research of the NELL-1 (搜索) protein. Development efforts for its bone graft substitute product focus on bone regeneration in spinal fusion procedures, while the company additionally holds rights to trauma and osteoporosis (搜索) applications.
