Clinical Testing Results
In a study published in the Chemical Engineering Journal, the sensor was tested on 51 people with type 2 diabetes (search) and 20 healthy volunteers. All participants exhaled into aluminum foil bags, and the sensor successfully distinguished between diabetic patients and healthy individuals based on their breath samples. The device demonstrated high sensitivity, detecting acetone (search) at concentrations as low as 4 parts per billion with response and recovery times of approximately 20 seconds.
"This sensor only requires that you exhale into a bag, dip the sensor in and wait a few minutes for results," said senior researcher Huanyu "Larry" Cheng, an associate professor of engineering science and mechanics at Penn State.
Importantly, the sensor response correlated with participants' blood sugar levels, suggesting potential applications beyond simple diabetes (search) diagnosis. "If we could better understand how acetone (search) levels in the breath change with diet and exercise, in the same way we see fluctuations in glucose levels depending on when and what a person eats, it would be a very exciting opportunity to use this for health applications beyond diagnosing diabetes," Cheng explained.
Technology and Design
The sensor utilizes laser-induced graphene (LIG) created by exposing polyimide film to a carbon dioxide laser. This process produces a porous graphene structure that is permeable to gases, increasing the likelihood of capturing acetone (search) molecules. To enhance selectivity for acetone, researchers incorporated zinc oxide, forming a junction that responds specifically to acetone while ignoring other gases.
A significant technical challenge was addressing the humidity of human breath, as water molecules can compete with acetone (search) for binding sites on the sensor. The research team solved this by adding a molecular sieve membrane that blocks water vapor while allowing acetone to pass through, enabling the device to function effectively in humid conditions.
Addressing Diagnostic Gaps
The technology targets a substantial unmet need in diabetes (search) detection. Of the 37 million adults with diabetes in the United States, approximately 20% remain undiagnosed. Current diagnostic methods typically require doctor visits, laboratory work, and blood draws, which can be costly or inconvenient barriers that prevent screening for people unaware they may have diabetes.
Diabetes (search) significantly increases the risk of complications including heart disease (search), stroke (search), kidney disease (search), and death. While most of these outcomes can be managed with proper treatment, the lack of diagnosis prevents timely intervention. A breath-based test could reach populations not served by existing diagnostic methods, creating opportunities for earlier diagnosis and treatment.
Future Development
The current prototype requires patients to exhale into a bag before analysis to avoid interference from ambient air. However, researchers are working to adapt the design for more convenient use, potentially allowing the sensor to be positioned directly under a person's nose or integrated into a face mask for continuous monitoring.
