Cambridge, Massachusetts-based Caldera Therapeutics (搜索) has emerged from stealth with $112.5 million in funding and announced that first subjects have been dosed in a Phase 1 trial of its lead program CLD-423 (搜索), a bispecific antibody designed to treat inflammatory bowel disease (搜索) (IBD).
The company completed a $75 million Series A round from founding investors Atlas Venture (搜索), LAV and venBio in April 2025, followed by a $37.5 million Series A-1 financing led by Omega Funds (搜索) with participation from Wellington Management and Janus Henderson Investors.
Novel Dual-Target Approach for IBD
CLD-423 (搜索) represents a first-in-class bispecific antibody that simultaneously targets two clinically validated pathways: IL-23p19 and TL1A (搜索). This dual-targeting strategy aims to address what Caldera describes as the "efficacy ceiling" that traditional IBD treatments targeting single proteins have encountered.

The drug targets IL-23p19, a subunit of IL-23 (搜索) that is already the target of widely used drugs such as AbbVie (搜索)'s Skyrizi, as well as TL1A (搜索), which has captured attention from major pharmaceutical companies including Sanofi and Merck (搜索) & Co.
Strategic Licensing from China
Caldera licensed CLD-423 (搜索) from China's Qyuns Therapeutics (搜索) in April for $10 million upfront, just under 25% of equity in Caldera, and potential milestone payments of up to $545 million. The company follows an increasingly popular strategy of partnering with Chinese biotechnology companies, which benefit from lower costs and more flexible regulatory environments that allow faster progression from launch to clinical trials.

Market Opportunity and Clinical Development
Caldera is targeting the IBD market, which analysts estimate will reach $30 billion by the end of the decade. The category includes treatments for ulcerative colitis (搜索) and Crohn's disease (搜索), both caused by inflammation in the digestive tract.
The company has initiated a Phase 1 healthy volunteer trial of CLD-423 (搜索), with the goal of enabling clinical evaluation in IBD patients. Caldera was founded in 2025 with a senior leadership team experienced in IBD therapeutic discovery and development, as well as a proven track record in building biotechnology companies through successful exits.
The company joins other well-financed biotechnology companies that have emerged in recent years built around therapies acquired from China, including Kailera Therapeutics, Ouro Medicines, Expedition Therapeutics, and Candid Therapeutics.
