The study, published by the Columbus, Ohio-based molecular pathology company, demonstrated 100% efficacy in treated mice that would otherwise develop and die from amyotrophic lateral sclerosis (search).
Preclinical Efficacy Results
The research showed that mice treated with venetoclax maintained their ability to move, avoided paralysis, and exhibited dramatically less damage to their nerves and brain tissue compared to untreated controls. Additionally, the drug reduced inflammation in the nervous system, a known contributor to ALS (search) progression.
"This is a major step forward," said Gerard Nuovo, MD, the lead investigator at Gnome Sciences (search). "We've shown that a drug already approved for cancer (search) can stop ALS (search) before it starts, in animals—and we see the same molecular signatures in human brain tissue. That's incredibly promising."
Human Tissue Analysis Reveals Target Mechanism
The research team's analysis of brain tissue from deceased ALS (search) patients revealed that BCL2 family proteins (search) were significantly more active in ALS-affected neurons compared to healthy ones. These proteins were found in the same areas as other known ALS markers, suggesting they may play a key role in disease pathogenesis and treatment response.
The BCL2 (search) proteins have important roles in key cellular pathways, including apoptosis, autophagy, mitophagy, and clearing of misfolded and aggregated proteins—all processes implicated in neurodegenerative diseases.
Clinical Translation Underway
Gnome Sciences (search) has initiated a clinical trial to evaluate venetoclax in human ALS (search) patients. "We have begun a clinical trial to evaluate the drug in human ALS," said Dr. A. Mikhail, CEO of Gnome Sciences. "We have guarded optimism, but it is too early at this stage to make clinical recommendations, especially because this class of drugs is not without risks."
