Cornell Scientists Achieve Breakthrough in Reversible Male Contraception Using Meiosis Disruption

Cornell scientists have achieved a major breakthrough toward developing a safe, reversible, long-acting and 100% effective nonhormonal male contraceptive by successfully targeting meiosis (search), the cellular process that produces sperm. The proof-of-principle study, conducted over six years in mice, demonstrates for the first time that interrupting a key step in sperm development can temporarily halt fertility without causing lasting harm.
The research, published April 7 in the Proceedings of the National Academy of Sciences, utilized JQ1, a small molecule inhibitor originally developed to study cancer and inflammatory diseases. While JQ1 itself is not suitable as a treatment due to neurological side effects, it effectively disrupts prophase 1 (search), a critical stage of meiosis (search), allowing researchers to prove that targeting this process can safely shut down sperm production.
"We're practically the only group that's pushing the idea that contraception targets in the testis are a feasible way to stop sperm production," said Paula Cohen, professor of genetics at Cornell's College of Veterinary Medicine and director of the Cornell Reproductive Sciences Center, who served as senior author of the study.
Addressing Critical Unmet Medical Need
Current male contraceptive options remain severely limited to condoms and vasectomies. Many men are reluctant to undergo vasectomies despite their technical reversibility through subsequent surgery. Meanwhile, researchers have been cautious about developing hormonal approaches due to safety concerns observed in women taking hormonal contraceptives.
"So we were really motivated to look for nonhormonal contraceptive targets in the testis, something that stops sperm production without affecting male libido and secondary sex characteristics," Cohen explained. These secondary characteristics include facial and chest hair, deep voice, and muscle mass.
Strategic Targeting of Meiosis
The Cornell team strategically chose to target meiosis (search) rather than other stages of sperm development to ensure complete cessation of sperm production while preserving the ability to recover fertility. Sperm production occurs in three main stages: stem cell proliferation and differentiation, meiosis (where cells divide from 46 to 23 chromosomes), and spermiogenesis (final sperm cell formation).
"We didn't want to impact the spermatogonial stem cells (search), because if you kill those, a man will never become fertile again," Cohen said. The researchers also avoided targeting later stages because "once sperm entered spermiogenesis, there was a potential for viable sperm to leak out and fertilize an egg."
Mechanism and Recovery Results
JQ1 works by disrupting meiosis (search) during prophase 1 (search), causing developing cells to die at that stage while blocking gene expression required for later sperm development stages. In the study, male mice received JQ1 treatment for three weeks, during which sperm production stopped completely and all molecular parameters of meiosis were disrupted, including chromosomal behavior during prophase 1.
The recovery results proved remarkable. Within six weeks after treatment cessation, most healthy parameters of prophase 1 (search) returned along with normal sperm production. The researchers then bred the treated mice and confirmed complete fertility restoration. Importantly, the offspring were also healthy and able to reproduce normally.
"Our study shows that mostly we recover normal meiosis (search) and complete sperm function, and more importantly, that the offspring are completely normal," Cohen said.
Future Development and Clinical Translation
Moving forward, Cohen's team is developing three new gene targets that completely eliminate meiosis (search) and sperm production while maintaining overall mouse health. The researchers are now working to demonstrate these candidates can be targeted reversibly.
Cohen and colleagues plan to launch a company within the next two years to continue developing these methods. A future male contraceptive would likely be administered as an injection every three months or possibly as a patch to ensure effectiveness.
The team is also considering targets that disrupt meiosis (search) even earlier, just at entry into prophase 1 (search), which would further ensure no sperm survive and improve drug delivery by avoiding the blood-testis barrier that protects later stages of sperm development.
