Critical Path Institute (搜索) announced the official launch of the Critical Path Disease Modeling Coalition (CP-DMC) on pediatric inflammatory bowel disease (搜索), marking a significant advancement in how medicines are developed for children with chronic inflammatory conditions. The multi-stakeholder, public-private initiative aims to develop regulatory-grade, quantitative disease progression and care models to accelerate and better inform drug development and evaluations for pediatric Crohn's disease (搜索) and ulcerative colitis (搜索).
Strategic Partnership Framework
The coalition brings together leading academics and clinicians from the European Society for Paediatric Gastroenterology Hepatology and Nutrition (搜索) (ESPGHAN), including Prof. Lissy de Ridder, M.D., Ph.D., Head of Department of Pediatrics at Leiden University Medical Center (搜索). The initiative is supported by the Crohn's & Colitis Foundation (搜索) and establishes Quinten Health (搜索) as the founding AI and disease modeling partner.

Advanced Modeling Capabilities
CP-DMC operates alongside C-Path's Quantitative Medicine Program and Data Collaboration Center to integrate clinical trial and real-world data at scale for model development and application. The models will transform integrated longitudinal datasets into actionable quantitative descriptions of disease trajectories, treatment response, and variability in pediatric populations.
These regulatory-grade models are designed to inform regulatory submissions and optimize trials, serving as reusable platforms that reduce patient burden and strengthen evidence generation across programs. The approach represents a shift toward more sophisticated, data-driven methodologies in pediatric drug development.
Addressing Critical Gaps in Pediatric Care

The project addresses significant unmet needs in pediatric IBD treatment development, where traditional approaches have often fallen short of capturing the complexity of disease progression in younger populations. By creating fit-for-purpose reference simulation models usable across programs and sponsors, the coalition aims to standardize and accelerate the development process.
Expansion and Future Applications
The CP-DMC Pediatric IBD Project serves as a proof-of-concept for the broader disease modeling coalition platform. Following successful pilots, CP-DMC will expand to support multi-indication portfolios, with an evolving structure designed to ensure sustainability, regulatory impact, and patient relevance.

C-Path is actively seeking expressions of interest from industry partners and data owners to contribute datasets and join the initiative. The organization, founded in 2005 as a public-private partnership in response to the FDA's Critical Path Initiative, currently includes more than 1,600 scientists and representatives from government and regulatory agencies, academia, patient organizations, disease foundations, and pharmaceutical and biotech companies.
