Curcumin Shows Best Balance of Anticancer Activity and Safety Against Fibrosarcoma Cells in Comparative Study

A new laboratory study from Wroclaw Medical University (search) and the University of Wroclaw (search) has identified curcumin as the plant-derived compound offering the most balanced profile of anticancer activity and safety against fibrosarcoma (search) cells. The research, published in the International Journal of Molecular Sciences, compared five natural substances—curcumin, berberine, biochanin A, cucurbitacin E (search), and CAPE (search) (caffeic acid phenethyl ester)—and found that strong tumor-killing potential does not always correlate with tolerability.
The study centered on the NF-κB (search) signaling pathway, a critical regulator of inflammation, metabolism, cell survival, and cellular aging. "NF-κB acts like a central control switch that integrates cellular stress signals and determines whether a cell activates inflammatory pathways, fights for survival or changes its metabolism," explained Prof. Julita Kulbacka from the Department of Molecular and Cellular Biology at Wroclaw Medical University (search). In cancer cells, excessive NF-κB activation can promote survival and adaptation to hostile conditions, yet the pathway remains essential for normal cell function, making precise modulation—rather than complete shutdown—the therapeutic goal.
Cancer cells suffer disproportionate energy depletion
All five compounds disrupted mitochondrial function, substantially reducing the cells' capacity to generate energy. In fibrosarcoma (search) cells, ATP levels decreased by approximately 83–92%, whereas healthy muscle cells experienced reductions ranging from only 23–73%. This disparity suggests that cancer cells are markedly more vulnerable to disturbances in energy metabolism.
The compounds also influenced mitophagy, the cellular machinery responsible for clearing damaged mitochondria. "Berberine, cucurbitacin E (search) and CAPE (search) enhanced PINK1/PARKIN-dependent mitophagy in fibrosarcoma (search) cells. Combined with the marked reduction in ATP levels, this indicates not a protective response but an overwhelmed adaptive mechanism leading to cellular senescence and death," said Prof. Kulbacka.
Curcumin drives strongest senescence response
Each of the five compounds induced features of cellular senescence—a state in which cells remain alive but permanently lose the ability to divide—in fibrosarcoma (search) cells. Curcumin produced the most pronounced effect: the proportion of senescent cancer cells rose from approximately 16.5% to more than 75%. The other compounds also pushed the senescent fraction above 66%. Healthy muscle cells generally showed a weaker response, pointing to a degree of selectivity toward malignant cells.
Cell viability experiments further demonstrated that curcumin and CAPE (search) exhibited the greatest selectivity against fibrosarcoma (search) cells. "Our findings show that these natural compounds can selectively place a greater metabolic burden on cancer cells than on healthy cells by disrupting mitochondrial function and energy metabolism. Among the compounds tested, curcumin demonstrated the most balanced profile," emphasized Prof. Kulbacka.
Safety testing reveals critical tradeoffs
To assess whole-organism tolerability, the researchers employed greater wax moth (Galleria mellonella) larvae as an initial toxicity model. Curcumin and berberine were the best-tolerated compounds. CAPE (search) and cucurbitacin E (search) caused higher mortality, while biochanin A proved the most toxic of the five substances tested. These findings underscore that anticancer potency alone is insufficient for identifying promising drug candidates—safety for healthy tissues and the entire organism carries equal weight.
A foundation for future research, not a current treatment
The researchers caution that the results do not mean curcumin, berberine, or any of the other tested compounds can currently be used to treat sarcomas in patients. The study was conducted using animal cell lines and a simple invertebrate model. "Our findings come from laboratory experiments and preliminary tests in wax moth larvae. The next step is to evaluate these compounds in more advanced in vivo models and to better understand their mechanisms of action, optimal dosing and delivery methods," noted Prof. Kulbacka.
