London-based Cytospire Therapeutics (搜索) announced Tuesday it has raised £61 million ($83 million) in Series A funding to advance its novel approach to cancer immunotherapy through pan-gamma delta T cell engagers. The oversubscribed round was led by 4BIO Capital (搜索), with participation from new investors including Servier Ventures (搜索), British Business Bank, Sound Bioventures, and Criteria Bio Ventures, alongside existing backers Abingworth (搜索) and LifeArc Ventures.
Targeting a Unique T Cell Subset
Cytospire is developing a new form of T cell engagers that specifically activate gamma delta T cells (搜索), a distinct subset of immune cells with unique anti-cancer properties. Unlike conventional T cell engagers that target the CD3 (搜索) receptor on all T cells, Cytospire's approach focuses on gamma delta T cells, which CEO Natalie Mount describes as being "already present in the immune-suppressing barrier, or 'tumor microenvironment,' cancers use to protect themselves."
According to Mount, gamma delta T cells (搜索) are "more fit to deliver an impact than other T cell types" and "can also better detect differences between target antigens on tumor cells and healthy ones." This selectivity could potentially address the efficacy and safety challenges that have limited conventional T cell engagers, particularly in solid tumor applications.
Lead Program Targets EGFR-Positive Cancers
The company's lead program, CYT X300 (搜索), is being developed for EGFR (搜索)-positive solid tumors, including colorectal, head and neck, and non-small cell lung cancers. The program is currently advancing through IND-enabling preclinical studies and GMP manufacturing, with plans for first-in-human clinical trials.
Cytospire's pan-gamma delta T cell engagers are "uniquely engineered to target all gamma delta T cells (搜索) rather than specific subtypes," according to the company. This approach aims to "overcome patient heterogeneity and generate broader, more effective anti-tumour immune responses through the activation of both tissue/tumour resident and blood resident effector cells."
Experienced Leadership and Strategic Vision
Mount brings significant experience in the T cell engager space, having previously led Adaptate Biotherapeutics and served as chief scientific officer of GammaDelta Therapeutics, both of which were acquired by Takeda Pharmaceutical (搜索). She has since joined Abingworth (搜索) as a venture partner, which led Cytospire's seed round.
"What we want to achieve with the engager is to provide something in a biologic format which is very familiar to big pharma and has the ability to be scaled to be widely available to large numbers of patients," Mount stated.

The funding positions Cytospire to advance its differentiated immunotherapy platform in a competitive landscape where multiple companies are developing next-generation T cell engagers to overcome the limitations of current approaches, particularly in solid tumor applications where conventional T cell engagers have faced significant challenges.
