Diakonos Oncology Secures $7 Million CPRIT Grant to Advance DOC1021 Dendritic Cell Therapy in Refractory Melanoma

Novel Dendritic Cell Platform Targets Treatment-Resistant Disease
DOC1021 represents a first-in-class, patient-derived double-loaded dendritic cell therapy that uniquely combines tumor lysate and amplified tumor-derived mRNA. The immunotherapy is manufactured using a patient's dendritic cells (搜索) combined with mRNA and proteins prepared from freshly obtained patient tumor specimens.
"Across more than two dozen preclinical tumor models and multiple clinical indications, our personalized dendritic cell vaccine platform has shown a consistent pattern of potent immune activation and efficacy," said Jay Hartenbach, President and COO of Diakonos Oncology. The unique double-loading approach mimics a viral infection, unlocking what the company describes as a synergistic and exponentially more powerful tumor killing TH1 response (搜索) driven by dual protein and RNA antigen sourcing. This mechanism allows targeting of the complete cancer antigen pool without requiring molecular modification of the patient's immune cells for manufacturing or preconditioning chemotherapy and high dose IL-2 for administration.
Addressing Critical Unmet Need in Refractory Melanoma
Refractory melanoma (搜索) represents a particularly challenging clinical scenario where the disease no longer responds to immune checkpoint inhibitors, resulting in limited treatment options and poor clinical outcomes. While many melanoma (搜索) cases can be treated effectively when detected early, some patients experience disease progression despite available therapies such as immune checkpoint inhibitors or targeted therapies.
"Despite the remarkable progress made with targeted therapies and immune checkpoint inhibitors, patients with refractory melanoma (搜索) still have very limited options," said Ryan J. Sullivan, MD, Director of the Cutaneous Medical Oncology Program at Massachusetts General Brigham Cancer Institute (搜索). "DOC1021's personalized dendritic cell approach, which presents each patient's own tumor antigens (搜索) in a viral-mimicking context, offers a compelling new way to re-engage the immune system against tumors that have escaped standard therapies."
Clinical Development Timeline and Study Design
The planned Phase 1/2 study will evaluate the safety and preliminary efficacy of DOC1021, along with ctDNA and immune biomarker responses, in patients with refractory melanoma (搜索). Enrollment in the Phase 1 clinical study is expected to begin in January 2026, representing the third target indication for DOC1021's clinical development program.
DOC1021's design allows for simple administration in the outpatient setting and broad reach via community cancer centers, potentially expanding access to this personalized immunotherapy approach.
Expanding Clinical Portfolio
Diakonos currently operates two active clinical trials with DOC1021: a Phase 1 pancreatic cancer (搜索) study (NCT04157127) and a Phase 2 glioblastoma (搜索) study (NCT06805305). The company has received Fast Track designations from the FDA for both the glioblastoma and pancreatic cancer programs, awarded in October 2023 and May 2024, respectively. Additionally, Diakonos received Orphan Drug Designation for the glioblastoma program in January 2024.
The refractory melanoma (搜索) Phase 1/2 study represents an important expansion of DOC1021's clinical development into a third indication, supported by the substantial CPRIT (搜索) funding that will facilitate the trial's initiation and execution.
