Editas Medicine Targets 2026 Patient Dosing for In Vivo CRISPR LDL Therapy EDIT-401

Editas Medicine says it remains on track to dose the first patients this year with EDIT-401 (搜索), an in vivo CRISPR therapy designed to lower LDL cholesterol by upregulating the LDL receptor.

Editas Medicine is advancing EDIT-401 (搜索), an in vivo CRISPR gene-editing therapy designed to lower LDL cholesterol by increasing LDL receptor levels, and says it remains on track to dose patients this year in a clinical study of the program. Speaking at the Cantor Healthcare Conference, President and Chief Executive Officer Gilmore O'Neill said the company has completed its shift toward developing in vivo therapeutics using CRISPR editing.
Editas is prioritizing programs designed to produce meaningful efficacy improvements, use mechanisms differentiated from other drug modalities, offer measurable biomarkers for rapid clinical decision-making, and potentially be delivered through a single infusion or injection.
Mimicking a Naturally Occurring Gain-of-Function Variant
EDIT-401 (搜索) is designed to reduce LDL cholesterol by increasing levels of the LDL receptor, which removes LDL cholesterol from circulation. O'Neill said the program uses CRISPR-Cas9 and CRISPR-Cas12 editing tools to mimic naturally occurring gain-of-function genetic variants rather than applying gene editing to mechanisms accessible through antibodies, antisense therapies or siRNA medicines.
The approach is based on a naturally occurring LDLR (搜索) variant identified in an Icelandic family whose members had low LDL cholesterol levels but were otherwise healthy, according to O'Neill. The variant involved a deletion in the three-prime untranslated region of the LDLR gene, removing microRNA binding sites and extending the messenger RNA's half-life, enabling production of more LDL receptor protein.
Editas selected a guide RNA pair that does not perfectly replicate the naturally occurring deletion but was more potent in exploratory work, O'Neill said. In non-human primate studies, EDIT-401 (搜索) produced a mean 90% reduction in LDL cholesterol, along with reductions in other atherogenic lipoproteins including lipoprotein(a) and ApoB. The company said it observed at least a sixfold increase in LDL receptor expression in the liver in those studies.
O'Neill contrasted the strategy with approaches targeting PCSK9, saying PCSK9 inhibition may face a ceiling effect because it prevents degradation of LDL receptors already produced by liver cells rather than directly increasing LDLR (搜索) production. He said reductions of roughly 50% to 60% with PCSK9-directed approaches have been transformative but that Editas aims to produce greater LDL lowering through a different mechanism.
Regulatory Submission in Australia and Phase I/II Design
Editas has submitted documentation to Australia's Human Research Ethics Committee, or HREC, for EDIT-401 (搜索) and is answering questions related to items such as the informed-consent form and starting dose. O'Neill said the company remains on schedule to dose patients this year, though he declined to specify the exact timing before receiving HREC clearance. If clearance were received, site activation could require a couple of months before the first patient is dosed, he said.
The planned Phase I/II study has two parts. Part one will use serial dose-escalating cohorts under a 3+3 design, with at least three patients per cohort and at least four cohorts. Editas is targeting a human-equivalent dose of about 0.6 milligrams per kilogram, which O'Neill said corresponds to the dose level expected to be fully efficacious based on non-human primate findings.
First-cohort safety data are expected in the first quarter of next year. Top-line safety and efficacy data across multiple dose cohorts are expected later in 2027. Part two would expand the number of patients treated at the selected dose, with enrollment anticipated to be completed by the end of 2028.
O'Neill said the company expects to review results from the first phase of the study with regulators as it considers a pivotal program. He said approvals based on LDL cholesterol outcomes have precedent in studies involving a couple hundred patients, although any such plan would depend on discussions with regulators.
Persistent Unmet Need Despite Existing Lipid-Lowering Therapies
O'Neill said a substantial number of high-risk patients remain above LDL cholesterol goals despite treatment with statins, inclisiran and other therapies. Citing Merck (搜索)'s Coral Reef study, he said 50% of patients receiving inclisiran alongside other cholesterol-lowering medicines did not achieve LDL cholesterol below 40 milligrams per deciliter. He estimated the potential U.S. population of patients with heterozygous familial hypercholesterolemia (搜索) and established cardiovascular disease (搜索) who remain above target at roughly 9 million to 10 million.
Financial Runway and Pipeline Priorities
Chief Financial Officer Amy Parison said Editas had $212 million in cash as of the second quarter and expects its capital to fund operations into the second half of 2028. She said the runway is intended to support advancement through the first part of the Phase I/II study and into part two, allowing the company to seek human proof of concept for EDIT-401 (搜索).
Editas also continues discovery-stage work on an in vivo hematopoietic stem cell-targeting program with potential applications in sickle cell disease (搜索) and thalassemia (搜索). However, O'Neill said the company's primary allocation of resources remains focused on advancing EDIT-401 (搜索).
