Eli Lilly Partners with Seamless Therapeutics in $1.1 Billion Gene Editing Deal for Hearing Loss

Eli Lilly and Company has entered into a worldwide strategic research and licensing agreement with Dresden biotech company Seamless Therapeutics (搜索) GmbH worth up to $1.1 billion to develop programmable, recombinase (搜索)-based gene therapies for selected forms of genetically caused hearing loss (搜索).
The collaboration combines Seamless's proprietary recombinase (搜索) platform with Lilly's extensive development and clinical expertise in genetic hearing disorders (搜索). Seamless contributes technology that enables precise and large-scale DNA (搜索) insertions and modifications independently of cellular DNA repair mechanisms, representing a key differentiator from the more widely used CRISPR/Cas technology developed by Prof Frank Buchholz at the Medical Faculty of TU Dresden.
Recombinase Technology Platform
Within the project, Seamless will develop tailored, site-specific recombinases designed to correct disease-causing mutations in selected target genes. The programmable recombinases are engineered enzymes designed to make large, precise changes to DNA (搜索) at specific locations without relying on the cell's own DNA repair pathway. Due to the multiple activity centers of recombinases, both cutting and the insertion of very long DNA fragments can be achieved with high precision.
"What convinced Lilly was that we were able to present data from animal studies," commented Albert Seymour, CEO of Seamless Therapeutics (搜索). "We are now truly at the point where we are moving beyond platform development and heading decisively towards the clinic."
Financial Terms and Strategic Implications
The agreement includes a guaranteed upfront payment as well as committed research and development funding. Seamless may receive payments of up to $1.1 billion, including development and commercial milestone payments, though tiered, sales-based royalties are not included. In return, Eli Lilly receives an exclusive license in the hearing loss (搜索) field to advance the candidates preclinically and clinically and to commercialize them.
Albert Seymour emphasized the strategic importance of the collaboration: "Lilly is invested in advancing novel treatment approaches for genetic diseases and shares our vision of bringing genetic medicines to patients who currently have limited treatment options. This collaboration is a validation of our gene editing platform and its broad disease-modifying potential."
Lilly's Genetic Medicine Strategy
The drugmaker has been steadily building a pipeline of genetic medicines for multiple diseases through acquisitions and partnerships, looking beyond its blockbuster weight-loss and diabetes drugs Zepbound and Mounjaro for growth. Lilly previously acquired $1.3 billion worth of Verve Therapeutics to develop gene-editing therapies for reducing high cholesterol in people with heart disease, and purchased Akouos in 2022 for $487 million to access its gene-therapy candidate for hearing loss (搜索), which showed hearing restoration in children in an early to mid-stage trial in 2024.
Future Development Plans
Seamless does not intend to remain solely a service provider and development partner for the pharmaceutical industry. The Dresden-based company also aims to advance its own recombinase (搜索) program in a self-selected indication, with data and details on the exact target expected to be presented later this year.
"Our task is to identify and nominate the most suitable recombinase (搜索) from several developed candidates for further development by Lilly. Lilly contributes the expertise in the genetics of hearing loss (搜索). This model also lends itself to further collaborations that fit our corporate strategy," Seymour explained.
The company has raised over $40 million and, including Lilly's upfront payment, is well funded to advance its first experimental drug to lab studies by the end of the year. Seymour noted that the company is open to similar partnerships beyond Lilly, positioning Seamless's 'recombinase (搜索) on demand' technology prominently in the gene editing landscape.
