For the remaining 80% of non-responders, new treatment strategies are urgently needed.
Clinical Progress and Sample Collection
The international consortium has made substantial progress across multiple key areas as it approaches the end of its second year. Vita-Salute San Raffaele University (search) and IRCCS San Raffaele Hospital (search) (OSR) are leading clinical activities and have performed comprehensive tumor sequencing to identify expressed mutations that power downstream neoepitope prediction.
Nearly all patient samples have been collected and sequenced, with researchers successfully identifying key genetic alterations that inform tumor neoantigen (search) prediction. PBMCs and tumor-infiltrating lymphocytes (TILs) from surgical tumor biopsies of non-responder patients have been acquired and banked at the OSR Biobank. The teams' expertise in experimental immunology is actively advancing work on TIL expansion, functional characterization, and assay execution across the program.
AI-Driven Neoantigen Discovery
Project partner Ardigen has refined its computational workflow ARDentify, an AI-based platform designed to prioritize tumor-specific and tumor-associated antigens using patient genomic data. The platform selects peptide candidates for future validation, building on Ardigen's expertise in multi-omics integration and AI-guided discovery workflows.
This computational approach represents a crucial step in personalizing immunotherapy (search) by identifying the specific neoantigens that could serve as targets for patient-specific T cell therapies.
Advanced T Cell Isolation Technology
PEPperPRINT (search) has advanced and standardized its T-Plex-Capture workflow to isolate tumor-neoantigen (search)-specific CD8+ T cells (search). The innovative platform uses color-coded microspheres conjugated with streptavidin to bind biotinylated HLA-I (search) allotypes in a peptide-tethered form loaded with individual peptides derived from neoepitope predictions.
Cytokine-capture antibodies are incorporated at defined ratios, enabling the T-Plex-Capture method to achieve high-specificity detection and isolation of antigen-specific T cells directly from TILs and PBMCs. Work continues to improve and standardize advanced assays for the isolation of patient-specific T cells, representing an important step toward clinical applicability.
