FDA Clears Quantum BioPharma's IND for Lucid-MS, a First-in-Class PAD2 Inhibitor, in Phase 2 Multiple Sclerosis Trial
核心洞察
The FDA has cleared Quantum BioPharma's Investigational New Drug application for Lucid-MS, allowing a randomized, double-blind, placebo-controlled Phase 2 trial in multiple sclerosis (搜索).
Lucid-MS is a patented, first-in-class non-covalent inhibitor of peptidyl arginine deiminase 2 (搜索) (PAD2) designed to target demyelination rather than broadly suppress the immune system.
Preclinical models showed Lucid-MS prevented and reversed myelin degradation, and the Phase 2 study will assess safety, tolerability and potential therapeutic effects in patients with MS.
The U.S. Food and Drug Administration (搜索) has cleared Quantum BioPharma Ltd. (搜索)'s Investigational New Drug application for Lucid-MS, permitting the company to advance its lead multiple sclerosis (搜索) candidate into a randomized, double-blind, placebo-controlled Phase 2 clinical trial. The clearance marks the transition of Lucid-MS from preclinical research into human testing in patients with MS.
Lucid-MS is a patented, first-in-class, non-covalent inhibitor of peptidyl arginine deiminase 2 (搜索) (PAD2). Rather than broadly suppressing the immune system, as many existing MS therapies do, the candidate is designed to target demyelination directly — the underlying mechanism of multiple sclerosis (搜索). According to Quantum BioPharma, Lucid-MS has demonstrated the potential to prevent and reverse myelin degradation in preclinical models.
The company said the FDA clearance represents a significant development milestone and that the clinical program could represent the first human evaluation of a drug candidate based on this specific mechanism of targeting myelin degradation. The planned Phase 2 study is expected to evaluate Lucid-MS in people living with MS and to provide clinical data on the candidate's safety, tolerability and potential therapeutic effects.
Targeting the Root Cause of Nerve Damage
Multiple sclerosis (搜索) is a chronic disease in which the immune system attacks the protective myelin sheath around nerve fibers. Quantum BioPharma is developing Lucid-MS with the goal of protecting and potentially repairing the myelin sheath, which is damaged during disease progression. The company notes that current MS treatments can slow disease progression but do not directly repair myelin or restore neurological function that has already been lost.
If successful, Lucid-MS could offer a new mechanism of action for MS patients, potentially addressing the root cause of nerve damage rather than only modulating the immune response. Approximately one million people in the United States and nearly 2.9 million people worldwide are estimated to live with multiple sclerosis (搜索), a condition for which there is currently no cure.
Lucid-MS is the lead drug candidate of Lucid Psycheceuticals Inc. (搜索), Quantum BioPharma's wholly owned subsidiary, which focuses on developing therapies for neurodegenerative and other challenging disorders.
Imaging Collaboration Aims to Measure Demyelination
Beyond the clinical program, Quantum BioPharma is collaborating with Massachusetts General Hospital and Harvard Medical School on research involving the [¹⁸F]3F4AP PET imaging tracer. The tracer is being evaluated as a method to visualize and measure demyelination, which could provide a valuable tool for assessing treatment effects in MS and other demyelinating conditions. Such imaging tools are described as essential for developing therapies that target remyelination and neuroprotection.
Broader Portfolio and Financial Position
Quantum BioPharma is building a portfolio of assets targeting neurodegenerative and metabolic disorders as well as alcohol misuse disorders. The company invented UNBUZZD (搜索)™, a product for alcohol misuse, and spun out its over-the-counter version to Unbuzzd Wellness Inc. (搜索) Quantum retains a 19.84% ownership interest in Unbuzzd Wellness Inc. as of March 31, 2026, and is eligible to receive royalties equal to 7% of sales until cumulative payments reach $250 million, after which the royalty rate decreases to 3% in perpetuity. Quantum also retains full rights to develop similar products for pharmaceutical and medical uses.
Following the announcement, QNTM was trading at $3.55, up 9.12%.
