Novel Localized Delivery Technology
Enolen (search) represents a first-of-its-kind treatment approach utilizing novel anti-androgen eluting implants containing enzalutamide, an FDA-approved prostate cancer compound. The technology leverages Alessa's proprietary local delivery platform, which delivers anti-androgens directly to diseased tissue in the prostate through drug-eluting implants approximately the size of a grain of rice.
According to the company, this localized delivery approach can help eliminate potential side effects associated with systemic anti-androgen and testosterone-lowering drugs, including sexual dysfunction, muscle mass loss, cognitive issues, metabolic syndrome, and cardiovascular events.
Preclinical and clinical studies demonstrate that Alessa's implant technology can deliver durable and continuous release of effective anti-cancer agents, achieving high local drug concentrations while minimizing potential negative side effects from systemic exposure. The implants are designed to provide two or more years of continuous drug elution directly to diseased tissue.

Ongoing Phase 1 Trial
Enolen (search) is currently being evaluated in a Phase 1 trial in the United States, assessing safety, tolerability, and preliminary efficacy for localized sustained delivery of enzalutamide into the prostate. The study plans to enroll up to 20 patients who are planning for radical prostatectomy.
Trial participants receive the drug-eluting Enolen (search) implants and undergo repeat MRI at 6 to 12 weeks following implantation, followed by standard of care radical prostatectomy. Primary endpoints include incidence of adverse events and measurement of pharmacokinetics. Secondary assessments include MRI changes, testosterone level changes at 6 to 12 weeks following implantation, and prostate-specific antigen level changes at 6 to 12 weeks following radical prostatectomy.
Alessa expects to present initial findings from the Phase 1 study in 2026, providing crucial data on the safety and efficacy of this novel localized delivery approach for early-stage prostate cancer treatment.
