FDA Official's Skepticism Triggers Disc Medicine Stock Decline Amid Bitopertin Review
核心洞察
Disc Medicine shares dropped 11.5% following reports that FDA biologics director Vinay Prasad expressed skepticism about bitopertin's efficacy for treating porphyria (搜索).
The drug is under review through the FDA's Commissioner's National Priority Voucher program, which aims to reduce assessment times to one to two months.
Wall Street analysts dismissed the concerns as overblown, maintaining positive ratings while noting Disc management was "blindsided" by the report.
Disc Medicine shares tumbled Friday after reports emerged that a senior FDA official has expressed doubts about the efficacy of bitopertin, the company's experimental treatment for the rare blood disorder porphyria (搜索) currently under expedited regulatory review.
The stock closed down 11.5% at $80.04 after falling as much as 21.7% during trading, following a STAT News report indicating that Vinay Prasad, director of the FDA's biologics center, has become personally involved in the agency's review of bitopertin and expressed skepticism about the drug's effectiveness.
Priority Voucher Program Under Scrutiny
Bitopertin is among 18 drugs currently being reviewed under the FDA's Commissioner's National Priority Voucher (CNPV) program, announced in June to reduce assessment times from the standard 10-12 months to just one to two months. However, the STAT report suggested that political pressure surrounding the program is creating "uncomfortable situations" for career scientists at the FDA.
Prasad reportedly intervened after the FDA team handling Disc's approval application requested additional time for review, despite the compressed timeline mandated by the CNPV scheme. Notably, as a small molecule drug, bitopertin falls outside Prasad's typical remit as biologics center director.
Management Response and Analyst Reactions
Disc executives held an emergency call with analysts Friday to address the situation. According to Truist Securities (搜索) analysts, company management was "blindsided" by the report and unable to provide definitive clarity on the situation.
"We are in uncharted territory," Truist analysts noted, describing the unprecedented nature of Prasad's potential influence over the review process. The analysts emphasized that while Prasad's involvement warrants caution, "the information available today is far too limited to draw a definitive conclusion on the potential outcome."
BMO Capital Markets (搜索) analysts reported that Disc executives confirmed they have had no direct interactions with Prasad, though they understand he participates in "tumor board"-style CNPV reviews. According to BMO, the FDA has not communicated any efficacy concerns to the company, and no advisory committee has been deemed necessary.
Wall Street Maintains Confidence
Despite the market reaction, several Wall Street firms dismissed the concerns as overblown. Jefferies (搜索) analyst Roger Song characterized the stock decline as a "stock overreaction," maintaining confidence in bitopertin's approval based on "robust efficacy and safety data and to-date review experience."
TD Cowen (搜索) maintained its 'Buy' rating, stating that skepticism about the program's path is overblown and that management remains confident. BMO Capital preserved its 'Outperform' rating with a $120 price target, noting that while some procedural questions have been raised by the FDA, "the tone and tenor of interactions with FDA remains upbeat and collaborative."
Drug Profile and Timeline
Disc Medicine submitted its application for bitopertin approval on September 29 for patients aged 12 years and older with erythropoietic protoporphyria (搜索) (EPP). The company has expressed hopes for potential launch by the end of January 2026, consistent with the CNPV program's accelerated timeline.
EPP is a rare, inherited metabolic disorder that causes extreme sensitivity to sunlight, leading to immediate, painful skin reactions including burning, tingling, redness, and swelling due to protoporphyrin buildup in red blood cells from an enzyme deficiency.
The company stated earlier this month that its application has been officially accepted for review on schedule, with management noting that "commercialization efforts have been accelerated significantly and we are well on our way to launch readiness."
