The company emerged with $50 million in initial funding from Flagship and five years of platform development behind it.
Modified DNA Platform Addresses Key Genetic Medicine Limitations
Serif's approach centers on what CEO and co-founder Jacob Rubens describes as "modified DNA (搜索)" medicines that combine the strengths of multiple genetic medicine approaches while mitigating their individual limitations. The platform addresses fundamental barriers that have prevented DNA from becoming a practical therapeutic modality outside of gene therapy (搜索), specifically its immunogenicity and inefficient access to the cell's nucleus.
"Instead of making proteins outside of the body and injecting those proteins into us, we can actually make any drug inside of us by just delivering the DNA sequence that codes for that drug," Rubens explained.
The Modified DNA (搜索) treatments consist of two key components: finely tuned instructions for therapeutic proteins and mRNA (搜索) sequences for "co-factors" that enhance cellular uptake and nuclear entry. These components are delivered using specially designed lipid nanoparticles (搜索) rather than the engineered viruses commonly used in gene therapy (搜索).
Technology Stack Enables Durable, Redosable Therapeutics
Serif has developed what it describes as a complete technology stack that includes Modified DNA (搜索) designed to minimize innate immunogenicity, co-delivered mRNA (搜索) co-factors that enhance nuclear entry and gene expression, optimized lipid nanoparticles (搜索) for redosing and targeted tissue delivery, AI-guided DNA sequence design for cell-specific programmable gene expression, and scalable manufacturing processes.
According to the company, this approach enables treatments that are programmable, scalable, durable, and redosable without altering the cell's genome—properties that could reduce production complexity and costs while expanding therapeutic applications.
The modifications Serif makes to DNA are designed to enable therapies to "durably and safely express genes without altering a cell's genome—something that can risk causing unintended health problems," the company stated. The use of lipid nanoparticles (搜索) instead of viral vectors is intended to enable multiple dosing and broader tissue targeting.
Preclinical Data and Development Focus
Serif plans to present preclinical data at an upcoming scientific meeting demonstrating tolerability in non-human primates and sustained gene expression with therapeutic effects following intravenous administration. While the company has not disclosed specific conditions it's targeting initially, it is focusing on rare genetic diseases (搜索) and immunology applications where Modified DNA (搜索)'s therapeutic properties offer clear advantages for addressing unmet clinical needs.
The company positions its platform as addressing limitations of existing approaches: unlike RNA therapies that provide only transient gene expression, gene therapies that cannot be redosed, and genome engineering strategies that rely on DNA integration, Modified DNA (搜索) aims to enable a durable, redosable, and potentially safer therapeutic profile.

The launch represents Flagship Pioneering (搜索)'s continued investment in genetic medicine platforms despite recent funding challenges in the cell and gene therapy (搜索) sector. "When a foundational layer of biology becomes engineerable, new therapeutic categories are born that can redefine the possibilities for medicine," said Noubar Afeyan, Flagship's CEO and co-founder of Serif.
Serif is the first company publicly launched by Flagship in 2026, following the firm's 2025 unveiling of AI drug discovery startups and partnerships between its portfolio companies and major pharmaceutical firms including Pfizer and GSK.
The company's name draws inspiration from typography, where serif strokes at the edges of letters affect how readers perceive words—much like how modifications to synthetic DNA can change cellular responses to medicines, according to Rubens.
