Forward Therapeutics Initiates Phase 1 Trial of FT2109, Second-Generation Oral TNF Inhibitor
核心洞察
Forward Therapeutics (搜索) has dosed the first participants in a Phase 1 healthy volunteer trial evaluating FT2109 (搜索), a next-generation oral small molecule TNF (搜索) signaling inhibitor for chronic inflammatory diseases (搜索).
FT2109 (搜索) is designed to selectively inhibit soluble TNF (搜索)-TNFR1 (搜索) signaling while preserving membrane TNF (搜索)-driven regulatory mechanisms, potentially overcoming limitations of injectable anti-TNF biologics.
The randomized, double-blind, placebo-controlled study will assess safety, tolerability, pharmacokinetics, and pharmacodynamics of single and multiple ascending doses in healthy adults.
Forward Therapeutics (搜索) has announced the initiation of dosing in its Phase 1 clinical trial evaluating FT2109 (搜索), a next-generation oral small molecule designed to inhibit TNF (搜索) signaling for the treatment of chronic inflammatory diseases (搜索). The biotechnology company, based in Palm Beach Gardens, Florida, dosed the first participants in the healthy volunteer study on October 30, 2025.
Novel Mechanism Targets Selective TNF Inhibition
FT2109 (搜索) represents a departure from traditional anti-TNF (搜索) biologics by selectively targeting soluble TNF (搜索) (sTNF)-TNFR1 (搜索) signaling pathways. The oral small molecule is engineered to bind to soluble TNF, a primary driver of chronic inflammatory diseases (搜索), while preserving membrane TNF-driven regulatory and host defense mechanisms.
This selective approach addresses several limitations associated with injectable anti-TNF (搜索) biologics, which have revolutionized inflammatory disease treatment but carry significant drawbacks. Current biologics require injections, present elevated risks of infections and demyelinating events, can trigger paradoxical inflammatory responses, and may lose efficacy over time due to anti-drug antibody formation.
"Forward is building a portfolio of potentially best-in-class oral TNF (搜索) signaling inhibitors grounded in pharmacologic precision and long-term safety," said Toufike Kanouni, Co-Founder and Chief Executive Officer of Forward Therapeutics (搜索). "With FT2109 (搜索) now in the clinic alongside FT751 (搜索), we're executing on that vision, advancing small molecules designed to selectively and predictably modulate inflammation."
Phase 1 Trial Design and Objectives
The Phase 1 clinical trial follows a randomized, double-blind, placebo-controlled design evaluating single and multiple ascending doses of FT2109 (搜索) in healthy adult volunteers. The study will comprehensively assess the drug's safety profile, tolerability, pharmacokinetics, and pharmacodynamics. Additional evaluations will examine food effects and pharmacokinetics in elderly populations.
FT2109 (搜索) joins Forward's lead asset FT751 (搜索), another TNF (搜索) signaling inhibitor currently in Phase 1 clinical development. Both compounds demonstrated potent anti-inflammatory activity in preclinical studies, along with promising oral bioavailability and favorable safety profiles that offer advantages in tolerability and dosing flexibility.
Differentiated Drug Discovery Platform
Forward Therapeutics (搜索) develops its pipeline using SWIFT™, a proprietary data-driven drug discovery engine designed to target high-potential, traditionally difficult-to-drug targets. The platform combines curated, drug-like libraries with sophisticated screening assays and structural insights to enable rapid identification and optimization of lead compounds.
The company's focus on oral small molecule therapies for inflammation-driven diseases positions it to address significant unmet medical needs in chronic inflammatory conditions. As external validation builds for the oral TNF (搜索) inhibition class, Forward believes that differentiated pharmacokinetic and safety profiles can deliver meaningful and durable therapeutic impact for patients requiring long-term inflammatory disease management.
