From Village Tragedy to UN Advisor: Abdoulaye Djimdé's Fight Against Malaria

When Abdoulaye Djimdé was 11 years old, his baby brother died from cerebral malaria (search) in rural Mali. More than five decades later, Djimdé has become one of Africa's most influential malaria scientists—and in March, he was appointed to the United Nations Secretary-General's Scientific Advisory Board (search), a body that provides independent, evidence-based guidance on science and technology for sustainable development and policymaking.
"We African scientists research malaria (search) because we live the problem," Djimdé says. "Having African voices represented in those discussions would bring into consideration the perspective of the African continent."
The appointment places Djimdé alongside leading experts in fields ranging from artificial intelligence and economics to ecology and medicine. He describes the role as that of a "scientific ambassador for Africa," one that examines the directions science and innovation are taking and considers how they could increasingly benefit humanity.
A career forged in the field
Djimdé serves as CAMES Professor of Parasitology and Mycology, Chief of the Molecular Epidemiology & Drug Resistance Unit, and Director of the Parasites and Microbes Research and Training Centre at the University of Science, Techniques and Technologies of Bamako (search), Mali—one of the world's most malaria (search)-burdened countries.
His approach to research is defined by a commitment to field-based science. Unlike many colleagues in the Global North, whom Djimdé says work with "pet parasites" that have spent decades adapting to laboratory conditions, his team tests drug candidates against parasites collected directly from patients.
"In the Global North, colleagues tend to work with what I call pet parasites—parasites that have been in the lab for 30 years, adapted to lab conditions, grown and regrown so many times that their features are sometimes different from what you find in patients today," Djimdé says.
A recent study from his lab compared how well 14 antimalarial drugs performed against Plasmodium falciparum (search) field isolates collected from three different sites. The team identified significant differences in drug responses linked to each collection site, findings that could inform region-specific treatment strategies (J. Antimicrob. Chemother.-Antimicrob. Cracking chloroquine resistance
Djimdé's path into malaria (search) research was not straightforward. Strong marks in mathematics and physics led him into pharmacy rather than medicine, graduating from the Ecole Nationale de Medecine et de Pharmacie of Bamako in 1988. A turning point came in 1993, when he volunteered to assist American researcher Christopher Plowe on a field test investigating chloroquine resistance.
As Plowe's first doctoral student at the University of Maryland, Baltimore, Djimdé worked with scientists at the National Institutes of Health to identify a mutation in the gene coding for the Plasmodium falciparum (search) chloroquine-resistant transporter, or PfCRT (search). The NIH team found that the mutated protein stopped chloroquine from entering the parasite's vacuole, where the drug would otherwise kill the organism (Mol. Cell 2000, DOI: 10.1016/S1097-2765(05)00077-8).
Djimdé's team then validated the mutation in the field in Mali, treating malaria (search) patients with chloroquine and screening surviving parasites for the mutation. Every resistant parasite carried it; every susceptible one did not (N. Engl. J. Med. 2001, DOI: 10.1056/NEJM200101253440403). "That was really the first validation of this point mutation in the field," Djimdé says.
The group subsequently developed an assay requiring only a dried blood spot on filter paper—no cold chain or specialist equipment needed—that could be mailed like a normal letter. The mutated gene became the most widely used molecular tool for tracking chloroquine resistance globally. The resulting paper has been cited 1,350 times.
Rewriting the genetic map of African malaria (search)
By the early 2000s, large international genomics consortia were generating population-level data on P. falciparum from across the world. A conclusion kept surfacing: African parasite populations showed low genetic diversity and were considered scientifically uninteresting—the negative control.
"We used to attend these meetings and think, That is not the full story. We knew that malaria (search) behaved differently in different parts of the continent," Djimdé says. "And we knew that must have been driven by genetic differences."
In 2013, Djimdé and a group of African scientists formally launched the Plasmodium Diversity Network Africa, now called the Pathogen Genomic Diversity Network Africa. The group assembled and analyzed parasite genes—all collected from the field—from 15 African countries.
The results overturned the consensus. Parasites from southern, central, eastern, and western Africa were all genetically distinct from each other. "It completely changed the way the field thought about and studied African parasites," Djimdé says. The network now brings together experts from 17 countries to investigate the genetic diversity of pathogens including SARS-CoV-2, malaria (search) parasites, and antimicrobial resistance.
Making Pyramax available for Africa
Djimdé also became involved in one of the most consequential problems in malaria (search) drug development. Pyramax, a combination drug, had shown strong efficacy even against resistant strains, but early trials flagged liver toxicity cases. As part of its approval in 2012, the European Medicines Agency restricted it to a single lifetime dose, pending more data.
"A drug that you can only use once in a lifetime is not a drug for Africa," Djimdé says, noting that children on the continent suffer several malaria (search) episodes per year.
Between 2011 and 2016, his team tracked patients across repeated infections, working with a West African clinical trials network, Pyramax's South Korean codeveloper, and the nonprofit Medicines for Malaria Venture (search). Each patient was followed for two years. After tracking more than 14,000 malaria (search) episodes, the researchers showed that Pyramax was both safe and effective with repeated use (Lancet 2018, DOI: 10.1016/S0140-6736(18)30291-5). The drug is now registered in more than 20 countries across sub-Saharan Africa.
A new antimalarial on the horizon
One of Djimdé's most recent achievements has been coordinating African participation in the KALUMA trial of ganaplacide plus lumefantrine (search) (GanLum), the first successful Phase III trial in decades of a new type of antimalarial drug. The trial was conducted through the West African Network for Clinical Trials of Antimalarial Drugs, which Djimdé coordinates.
"It will be the first really new kind of antimalarial drug to reach the market in the last 25 years or so," Djimdé says. "We are very excited about it, especially amid growing concern about artemisinin resistance in sub-Saharan Africa. It will bring much-needed therapeutic novelty to the management of malaria (search)."
The treatment is now moving through regulatory review and could become available within the next two years. Djimdé notes that the international community, the drug manufacturer, and stakeholders are working to make it affordable to those who need it most.
Building the next generation
During the COVID-19 pandemic, Djimdé established the Pathogens Genomics Institute in Mali to strengthen local capacity in genomics and train young scientists from across Africa. He works with initiatives including DELTAS Africa and the Calestous Juma Science Leadership Fellowship to support emerging researchers in genomics, bioinformatics, and data science.
"We train them to master genomics, bioinformatics and big data science, to become much better at it than we are," he says. "The young folks' talent and energy make me hopeful that the continent is on the right path."
Djimdé's commitment extends beyond the laboratory. As a member of Rotary International, he has used grant-writing skills developed through science to secure funding for development projects in Pongonon, the village where he grew up—helping bring running water, an automated mill, and medical supplies to the community, which has grown from around 400 to nearly 2,000 inhabitants.
His long-term dream remains ambitious: a single pill capable of both preventing and treating malaria (search). "It's up to African scientists to tackle our problems, to take the lead and solve the continent's problems through scientific research," Djimdé says. "We need to face our difficulties head-on."
