The announcement comes against a backdrop of alarming statistics from the recent WHO GLASS Report, which revealed that approximately one in six laboratory-confirmed bacterial infections are now caused by antibiotic-resistant bacteria. Annual deaths associated with AMR are predicted to surge by 74.5%, rising from 4.71 million in 2021 to 8.22 million by 2050.
Targeting Critical Pathogens with AI-Driven Discovery
The six "Grand Challenges" programs, set to begin in early 2026 with full funding for three years, address some of the most formidable obstacles in antimicrobial research. The first initiative tackles Gram-negative bacteria, including E. coli (search) and Klebsiella pneumoniae (search), which pose particular challenges due to their complex cell envelope (search) defense systems and efflux pumps (search) that prevent antibiotic accumulation.
"Turning the power of supercomputers on the superbugs," researchers at Imperial's Drug Discovery Hub will partner with GSK scientists and Agilent Technologies (search) to generate novel datasets on diverse molecules. These efforts aim to create AI and machine learning models that enhance the ability to design antibiotics (search) for multi-drug-resistant Gram-negative infections, with all data and models made freely available to scientists worldwide.
A second program focuses on fungal infections (search), beginning with Aspergillus, which causes approximately two million infections annually with mortality rates exceeding 46% in high-risk ICU patients. With only four types of antifungal drugs (search) currently available, typically targeting the same cellular sites, this new initiative will use AI to identify unique fungal vulnerabilities for targeted drug development.
Advancing Vaccine Development Through Immune Modeling
The third major challenge addresses Staphylococcus aureus (search), one of the world's most dangerous drug-resistant pathogens responsible for more than one million deaths annually. Previous vaccine attempts against this pathogen have failed in clinical trials due to insufficient human-relevant data on bacterial behavior and immune responses.
The new research team will replicate surgical site infections under strictly controlled conditions to provide crucial data on infection progression and human immune responses to S. aureus, informing future vaccine development strategies.
Comprehensive Approach to AMR Prevention
The remaining three programs encompass disease surveillance using AI models to predict pathogen emergence and spread, innovative clinical trials to optimize antibiotic prescribing practices, and policy-focused research to inform public engagement and preventative interventions.

The collaboration will fund approximately 50 dedicated, multi-disciplinary scientific, clinical and academic roles, primarily based at Imperial College London facilities. This investment addresses a critical workforce shortage in AMR research, as highlighted by the AMR Industry Alliance report "Leaving the Lab" in 2024, which documented the field's struggle to attract necessary scientific expertise.
Global Impact and Open Science Approach
The Fleming Initiative (search), established by Imperial College London and Imperial College Healthcare NHS Trust, brings together research scientists, policymakers, clinicians, behavioral experts, and commercial partners to provide equitable AMR solutions at a global scale. GSK became the Initiative's first founding partner in 2024, pledging the £45 million investment.
