Trial Results Fall Short of Expectations
The September 2024 data readout showed 11.9uM of alpha-1 antitrypsin (AAT (search)) protein in patients treated with repeat injections of a 200-mg dose of WVE-006. While Wave characterized this outcome as "therapeutically relevant," the results failed to meet analyst expectations of 12uM and above, coming in "exceedingly close" but still below buyside projections, according to Truist Securities (search) analysts.
The underwhelming results represent a critical test for RNA editing technology, which is seen as a potentially safer and more flexible alternative to DNA editing. WVE-006 was the first treatment of its kind to reach human testing, making it an important barometer for the technology's capabilities across the industry.
Strategic Realignment and Future Plans
Wave framed GSK's decision as an opportunity to accelerate development more suited to the biotech's capabilities in rare diseases. "WVE-006 is well-suited to Wave's strengths and ability to execute on a commercial strategy. We look forward to engaging with regulators on how to rapidly advance this potentially transformative, first-in-class therapy," said CEO Paul Bolno.
Truist analysts noted that GSK's focus on "large indications, such as COPD (search)" may have influenced the decision, suggesting "AATD (search) may be a more niche program for them." The move comes as GSK's respiratory portfolio concentrates on "large-scale diseases" rather than rare conditions.
Upcoming Data and Regulatory Strategy
Despite the setback, Wave remains optimistic about higher dose cohorts. The company expects data for a 400-mg dose in the first quarter of 2026, which Truist analysts believe "could drive total AAT (search) well into the 13uM+ range." Additional findings from a 600-mg single dose and multidose cohorts are expected later this year.
Wave is "accelerating" its submission plans for WVE-006 and plans to meet with U.S. regulators to discuss accelerated approval requirements, with feedback expected in the middle of 2025. The company noted that GSK's decision was made before the next cut of clinical data became available.
Broader Collaboration Continues
Despite returning WVE-006, GSK maintains its broader partnership with Wave, which began in December 2022 with $170 million upfront and up to $3.3 billion in potential milestones. The collaboration provides GSK access to Wave's PRISM platform for RNA silencing and editing across up to eight preclinical programs.
"Our research collaboration with Wave continues with exciting opportunities ahead," said Tony Wood, GSK's chief scientific officer. In January, the companies selected a fourth program to advance under their deal, with Wave remaining eligible to receive up to $2.8 billion in initiation, development, launch and commercialization milestones, plus tiered royalties.
Financial Position and Market Context
Wave reported $196.2 million in cash and equivalents as of September 30, 2025, providing a runway into the third quarter of 2028. The company's financial stability comes amid broader challenges for RNA editing approaches, with rival Korro Bio having scrapped its own RNA editing therapy for AATD (search) after disappointing potency results.
Leerink Partners (search) analyst Joseph Schwartz suggested the move may be "at least partially informed" by recent developments from competitor Beam Therapeutics, which announced regulatory alignment on a speedy approval pathway for its gene editing treatment for AATD (search). The analyst also noted that GSK's new leadership under Luke Miels could represent "a new CEO sharpening and re-directing strategy."
