The compound, known as 4-nitro-substituted benzylic diselenide (search) (diselenide 7 (search)), targets critical survival pathways in cancer cells and has demonstrated strong anticancer activity in both laboratory experiments and animal models.
Multi-Pathway Targeting Approach
The research team, led by Dr. Asis Bala at IASST (search) and Dr. Krishna P. Bhabak from the Department of Chemistry at IIT Guwahati (search), synthesized the compound through nucleophilic substitution of benzylic halides with sodium-selenium derivatives. The compound attacks cancer through several mechanisms simultaneously, including inhibiting Akt/mTOR (search) and ERK (search) pathways that are critical for cancer cell survival, generating reactive oxygen species (ROS) that damage DNA and induce cancer cell death, and reducing inflammation by suppressing pro-inflammatory signaling that normally helps tumors thrive.
Promising Preclinical Results
In preclinical studies conducted in Swiss albino mice bearing breast adenocarcinoma (search), the compound demonstrated remarkable efficacy. Results showed significant reduction in tumor volume, inhibition of angiogenesis and metastasis, and prolonged survival compared with untreated controls. The findings, published in the Journal of Medicinal Chemistry, highlight the compound's ability to disrupt several key processes that help cancer survive, including tumor growth, blood vessel development, and the spread of cancer to other parts of the body.
Addressing Critical Medical Need
Triple-negative breast cancer (search) represents a significant clinical challenge, accounting for about 10-15% of all breast cancer cases and disproportionately affecting younger women, particularly those under 40, as well as individuals with BRCA1 mutations. The aggressive nature of TNBC (search) is reflected in its survival statistics: while localized TNBC shows approximately 91% five-year survival rates, regional spread drops to 65-66% survival, and distant metastatic disease has survival rates as low as 12%.
