Ipsen Unveils Corabotase, First-in-Class Recombinant Neuroinhibitor for Aesthetic Applications
核心洞察
Ipsen announced corabotase (搜索) (IPN10200), the first investigational molecule in a newly designated class of recombinant neuroinhibitors (RNI™) recognized by WHO and USAN.
Phase II LANTIC trial data demonstrated positive results for corabotase (搜索) in treating glabellar lines (搜索), showing rapid onset of action and sustained duration of effect.
The engineered molecule features optimized catalytic and binding domains designed to enhance receptor affinity, uptake, and resistance to degradation.
Ipsen has announced the development of corabotase (搜索) (IPN10200), marking a significant advancement in aesthetic medicine as the first investigational molecule in a newly designated class of recombinant neuroinhibitors (RNI™). The World Health Organization and United States Adopted Names Council have officially recognized this novel therapeutic class, highlighting the scientific innovation behind the compound.
The company presented Phase II data from the LANTIC trial at the SCALE 2026 symposium on May 16, 2026, demonstrating corabotase (搜索)'s efficacy in treating glabellar (frown) lines. The results, initially reported in September 2025, confirmed a differentiated clinical profile with rapid onset of action and clinically sustained duration of effect.
Advanced Protein Engineering
Corabotase (搜索) represents a breakthrough in recombinant protein engineering, designed through Ipsen's proprietary manufacturing platform. The molecule combines an active catalytic domain A with an affinity-enhanced binding domain B, with every structural element deliberately optimized to increase receptor affinity, enhance uptake, and improve resistance to degradation.
"Corabotase (搜索) has been designed with scientific rationale," said Christelle Huguet, PhD, Executive Vice President and Head of R&D at Ipsen. "Through advanced recombinant protein engineering and proprietary manufacturing capabilities, we have created a fully custom-designed molecule that brings an active catalytic domain A together with an affinity enhanced binding domain B. The recognition of corabotase as the first molecule in a new therapeutic class reflects both its scientific novelty and Ipsen's long-standing leadership in neuroscience."
This engineering approach enables long-lasting inhibition of neurotransmitter release (搜索) and sustained reduction in muscle activity, potentially offering advantages over existing treatments in the aesthetic medicine market.
Comprehensive Clinical Development Program
The LANTIC trial (n=727) is a Phase I/II study evaluating corabotase (搜索)'s safety and efficacy across three aesthetic indications for moderate to severe upper facial lines: glabellar lines (搜索), forehead lines (搜索), and lateral canthal lines (搜索). The trial is structured across three stages, with Stage 1 focusing on dose-finding and escalation in glabellar lines.
Stage 1 included three defined steps: dose-escalation (Phase Ib), dose-finding versus placebo and Dysport (Phase II), and additional dose-finding versus placebo and Dysport (Phase II). Step 3 provided the proof-of-concept data for corabotase (搜索) in glabellar lines (搜索), involving 183 patients. Stages 2 and 3 will evaluate corabotase across all three upper facial indications versus placebo.
The trial remains ongoing, with proof-of-concept data expected later this year for forehead lines (搜索) and lateral canthal lines (搜索). The LANTIC study represents one component of Ipsen's broader corabotase (搜索) development program, which includes multiple Phase II and Phase III trials advancing the compound across various aesthetic and therapeutic indications.
