The study will assess safety, tolerability, and signal finding in 75 patients with Parkinson's disease who experience apathy across 16 sites in Germany, Bulgaria, Poland, and Spain.
The trial is fully funded by IRLAB's development collaborator, the McQuade Center for Strategic Research and Development (搜索) (MSRD (搜索)), an indirect subsidiary of Otsuka Pharmaceutical Co., Ltd. The collaboration, established in May 2024, aims to advance IRL757 through proof-of-concept trials as a potential treatment for apathy (搜索).
Addressing a Critical Unmet Medical Need
Apathy (搜索) represents a substantial unmet medical need in Parkinson's disease (搜索), characterized by indifference, resignation, and lack of response to external stimuli. The condition affects a substantial proportion of people living with Parkinson's disease and leads to significant disability. Currently, no drugs are available on the market to treat apathy.

The prevalence of apathy (搜索) is substantial, occurring in 1.1-4 million people (20-70%) being treated for Parkinson's disease (搜索) in the eight major markets including China, EU5, Japan, and the United States. The condition also affects 4.9 to 6.7 million people (43-59%) being treated for Alzheimer's disease (搜索) in major markets such as Canada, China, EU, Japan, South Korea, and the United States. Overall, apathy affects over 20 million people in the United States and Europe alone without a currently available treatment.
Strong Safety Profile from Previous Studies
Two previous Phase I studies have demonstrated IRL757's favorable safety and pharmacokinetic profile. One study in healthy young adults and another in healthy older adults aged 65 and above both showed that IRL757 was well absorbed and provided good systemic exposure after 10 days of dosing. All participants completed both studies, and no serious adverse events were recorded.
Novel Mechanism of Action
IRL757 has shown positive effects in several preclinical models of cognitive function, including improved motivation. The drug candidate's efficacy is believed to be associated with its ability to counteract disturbances in central nervous system (搜索) nerve signaling proposed to underlie apathy (搜索) in several neurological conditions.
The compound has the potential to become the first treatment for apathy (搜索), with its efficacy thought to be linked to its unique ability to reverse disruption in cortical to sub-cortical nerve signaling, a key factor believed to contribute to apathy in neurological disorders.
