J&J Acquires Firefly Bio in $1 Billion Deal to Advance Pan-KRAS Degrader-Antibody Conjugates

Johnson & Johnson has agreed to acquire South San Francisco-based Firefly Bio (搜索) in an all-cash deal valued at $1 billion, the company announced Monday, marking a significant expansion of its oncology pipeline targeting KRAS (搜索)-driven cancers. The acquisition centers on Firefly's proprietary Firelink degrader-antibody conjugate (DAC) platform, a technology designed to overcome the limitations of existing treatments for tumors that have long resisted conventional therapeutic approaches.
"KRAS (搜索) has notoriously been considered an undruggable target and patients with KRAS-driven cancers continue to face limited treatment options with survival measured in months, not years," said John Reed, J&J's executive vice president of innovative medicine R&D. "We believe the proprietary Firelink platform will overcome the limitations of current treatments and diversify our pipeline with preclinical candidates for treating multiple types of solid tumours."
A Precision Approach to an Elusive Target
The Firelink DAC platform works by delivering highly selective protein degraders directly into cancer cells by conjugating them to antibodies, a strategy intended to spare healthy tissue from the collateral damage typical of existing treatments. The platform is directed at tumors driven by mutations in the KRAS (搜索) gene, which occur in nearly a quarter of all human cancers, with the G12D variant being the most commonly encountered.
Firefly Bio (搜索) emerged from stealth just over two years ago with $94 million in first-round financing backed by investors including Versant Ventures, MPM, and Eli Lilly. Since then, the company has been operating quietly while advancing its platform toward KRAS (搜索)-driven tumors, which have become a major focus for cancer drug developers.
The Growing KRAS (搜索) Landscape
J&J is already active in the KRAS (搜索) space through its partnership with Jacobio Pharma (搜索) on glecirasib, an oral KRAS G12C inhibitor currently heading for phase 3 clinical trials in patients with non-small cell lung cancer (搜索) (NSCLC) and colorectal cancer (搜索). Glecirasib belongs to the same class as the only two approved KRAS inhibitors to date — Amgen's Lumakras (sotorasib) and Bristol Myers Squibb (搜索)'s Krazati (adagrasib) — and is among a series of next-generation candidates advancing through the industry pipeline, including Eli Lilly's olomorasib, Genfleet's fulzerasib, and InxMed's garsorasib.
The pan-KRAS (搜索) class gained significant attention at this year's ASCO meeting, where Revolution Medicines' daraxonrasib — a related pan-RAS molecule — delivered a notable result in pancreatic cancer (搜索), a disease in which approximately 90% of cases harbor a KRAS mutation. The RASolute 302 study demonstrated unprecedented survival improvement in this notoriously aggressive and hard-to-treat malignancy.
Degraders Enter the Clinic
Several KRAS (搜索)-directed degraders have already entered clinical testing. Astellas Pharma's KRAS G12D-targeted ASP4396 reached phase 1 before being discontinued, while its follow-up candidate setidegrasib (ASP3082) is currently in phase 3 for NSCLC and pancreatic cancer (搜索). Arvinas is also advancing ARV-806, another G12D-targeted degrader, in phase 1 studies.
J&J said the Firefly acquisition expands its position in pan-KRAS (搜索) molecules designed to target all variants of the KRAS protein. The deal is expected to close before the end of 2026, pending regulatory approvals and other standard closing conditions.
