The study, led by Prof. Eijiro Miyako and published in Gut Microbes, focused on amphibian and reptile gut microbes due to the remarkably low incidence of spontaneous tumors in these animals. Despite living in pathogen-rich environments and enduring extreme cellular stress through metamorphosis and regeneration, these creatures maintain long lifespans relative to their size and rarely develop cancer unless exposed to direct environmental pollutants.
Bacterial Screening and Selection Process
The research team isolated 45 bacterial strains from Japanese tree frogs, Japanese fire belly newts (Cynops pyrrhogaster), and Japanese grass lizards (Takydromus tachydromoides). Through intensive screening, nine microbes demonstrated anti-tumor effects, with Ewingella americana (search) from tree frogs exhibiting the strongest response.
"Ewingella americana (search) exhibited remarkably potent cytotoxic activity with selective tumor-targeting ability characteristic of facultative anaerobic bacteria," the researchers noted in their findings.
Dual-Action Mechanism Against Cancer
The bacterium operates through a sophisticated two-pronged approach to eliminate tumors. E. americana demonstrates a natural affinity for the low-oxygen environment inside solid tumors, where it rapidly proliferates. Within 24 hours of administration, bacterial numbers increased approximately 3,000-fold specifically within tumor tissue, without affecting healthy organs.
The bacterium directly kills cancer cells through toxins secreted inside the tumor while simultaneously triggering a robust immune response. The bacterial invasion floods tumors with immune cells, particularly neutrophils, T cells, and B cells, alongside increases in inflammatory signaling molecules including TNF-α (search) and IFN-γ (search).
Superior Efficacy Compared to Standard Treatments
In mouse models of colorectal cancer (search), treatment with E. americana significantly outperformed standard therapies, including anti-PD-L1 antibody (search) and doxorubicin, in tumor regression studies. Every treated animal achieved complete tumor elimination with the single-dose treatment.
The response provided long-lasting protection beyond initial tumor clearance. When mice were later re-exposed to cancer cells, none developed new tumors, indicating the treatment had triggered durable immune memory against the cancer.
Safety Profile and Tumor Specificity
Comprehensive safety evaluations demonstrated minimal pathogenicity and no significant adverse effects at therapeutically effective doses. The bacterium was rapidly cleared from the bloodstream, becoming undetectable within 24 hours, and caused only short-lived inflammatory effects that resolved within 72 hours.
Over two months of observation, treated mice showed no signs of organ damage or chronic toxicity. The bacterium's selectivity for tumors arises from unique tumor characteristics including insufficient oxygen (hypoxia), leaky blood vessels, altered metabolism, and locally suppressed immune defenses.
Importantly, E. americana remained sensitive to antibiotics, providing intervention options if complications were to arise following treatment, contrasting favorably with genetically modified bacterial therapeutics.
Future Research Directions
The researchers plan to investigate E. americana's efficacy against breast cancer (search), pancreatic cancer (search), melanoma (search), and other malignancies while optimizing delivery methods and dosing protocols. They are also exploring potential combinations with existing immunotherapy and chemotherapy treatments.
"The successful identification of E. americana as a potent, naturally occurring anticancer agent establishes a proof-of-concept for microbiome-derived bacterial therapeutics and provides a foundation for the development of a new class of cancer treatments," the researchers concluded. "These discoveries may ultimately lead to transformative advances in precision oncology and offer new hope for patients with treatment-refractory cancers."
The findings suggest that gut microbiomes of lower vertebrates harbor numerous uncharacterized bacterial species with exceptional therapeutic potential, underscoring the critical importance of microbial biodiversity in advancing cancer treatment strategies.
