Levicept Ltd (search) announced that its novel neurotrophin-3 inhibitor LEVI-04 demonstrated significant structural and symptomatic benefits in a large Phase II osteoarthritis trial, with new data showing dose-dependent reductions in bone marrow lesions alongside pain relief. The findings will be presented at the American College of Rheumatology's ACR Convergence 2025 meeting in Chicago from October 24-29.
Phase II Trial Results Show Dual Mechanism Benefits
The multiarm, multicentre, randomized, double-blind, placebo-controlled Phase II study enrolled 518 participants with pain and disability due to knee osteoarthritis (ClinicalTrials.gov ID: NCT05618782). The trial revealed a significant, dose-dependent reduction in bone marrow lesion presence and area for LEVI-04 compared with placebo in patients with osteoarthritis.
Importantly, changes in bone marrow lesion area were significantly correlated with changes in patient-reported symptoms including pain, suggesting a direct relationship between structural joint improvements and clinical benefits.
Professor Philip Conaghan, MBBS, PhD, Director NIHR Leeds Biomedical Research Centre and Principal Investigator, emphasized the significance of these findings: "For the first time, we will show findings which suggest that LEVI-04 holds promise as a therapy to provide contemporaneous modification of joint structure - bone marrow lesions - and symptoms of OA including pain."
First-in-Class Neurotrophin-3 Inhibitor
LEVI-04 represents a first-in-class novel biologic under development as a non-addictive pain treatment. The drug works by inhibiting NT-3, one of the neurotrophin family of proteins. Previously presented data from the same trial showed that the primary efficacy endpoint was met with significant analgesia across all doses, accompanied by a favorable safety and tolerability profile.
