Eli Lilly and Company has acquired 4E Therapeutics (搜索), an Austin-based neuroscience company developing next-generation, non-opioid treatments for chronic pain (搜索), the companies announced Tuesday. The acquisition brings together 4E's innovative pipeline of orally available MNK (搜索) inhibitors with Lilly's broad neuroscience capabilities and global commercial infrastructure.
The deal marks Lilly's second acquisition of a non-opioid pain drugmaker in as many years, following its May 2025 purchase of SiteOne Therapeutics (搜索) in a transaction valued at up to $1 billion. Financial terms of the 4E acquisition were not disclosed.
Targeting the MNK (搜索)-eIF4E (搜索) Pathway in Peripheral Neurons
4E Therapeutics (搜索) has built its pipeline around a novel mechanism of action: inhibiting MNK (搜索) (MAP kinase-interacting kinase) enzymes to modulate the MNK-eIF4E (搜索) signaling pathway specifically in peripheral sensory neurons. This approach is designed to deliver meaningful pain relief while avoiding the central nervous system effects associated with many existing pain therapeutics, including the addiction risks linked to opioid-based treatments.
The company's lead compound, 4ET1103 (搜索), holds the distinction of being the first MNK (搜索) inhibitor developed to treat pain to advance to human clinical trials. In a Phase 1 study, 4ET1103 demonstrated a favorable safety profile. The asset is currently in development for nerve damage-related pain.
Beyond 4ET1103 (搜索), 4E's portfolio includes experimental treatments targeting migraines and acute pain, broadening the potential applications of its MNK (搜索) inhibition platform.
From Academic Research to Clinical Development

The company raised just under $10 million through private funding and grants from the National Institutes of Health to advance its research.

However, the path forward in pain drug development remains challenging. Lilly has discontinued two pain drug candidates from its pipeline within the same timeframe as these acquisitions, underscoring the inherent difficulties of advancing novel analgesics through clinical development.
