Lilly Breaks Ground on $6.5B Houston API Site to Manufacture Oral GLP-1 Foundayo

Eli Lilly broke ground on a $6.5 billion Houston, Texas facility that will produce active pharmaceutical ingredients for Foundayo (搜索) (orforglipron), its oral GLP-1 obesity (搜索) medicine.
The Generation Park site, expected to be operational by 2030, is the fifth Lilly API plant announced since 2020 under a $50 billion U.S. manufacturing push.
Lilly CEO David Ricks said roughly one-third of new GLP-1 pill patients are taking Foundayo (搜索), which entered the U.S. market in April and booked $98 million in second-quarter sales.
Eli Lilly and Company broke ground on a $6.5 billion manufacturing site in Houston, Texas, that will produce active pharmaceutical ingredients (API) for Foundayo (搜索) (orforglipron), the company's first synthetic oral GLP-1 medicine, along with other small-molecule medicines and advanced therapeutics.
The Houston facility at Generation Park is the fifth Lilly site designated to produce domestic API and one of ten U.S. manufacturing sites the company has announced since 2020. Lilly said it expects the plant to be operational by 2030. The groundbreaking comes nearly a year after the site was first announced, part of a $50 billion commitment to reshore medicine production.
Foundayo Manufacturing and Market Position
Foundayo (搜索) is a once-daily small-molecule (non-peptide) oral glucagon-like peptide-1 receptor agonist approved in the U.S. for weight management in adults with obesity (搜索), or adults with overweight who also have a weight-related condition, alongside a reduced-calorie diet and increased physical activity. It can be taken at any time of day with no planning around food or drink. Orforglipron was discovered by Chugai Pharmaceutical Co., Ltd. and licensed by Lilly in 2018.
Lilly noted that small molecules are manufactured using more well-established technologies and therefore can be more readily scalable than peptides. Foundayo (搜索) is currently under regulatory review in more than 40 countries, with a full global rollout targeted for 2027. The company is also studying the drug for type 2 diabetes (搜索), obstructive sleep apnea (搜索), osteoarthritis knee pain, hypertension, peripheral artery disease and stress urinary incontinence, with data expected later this year.
Speaking to CNBC from the Houston site, Lilly Chair and CEO David A. Ricks said about one-third of new GLP-1 pill patients are taking Foundayo (搜索), and that the company's share of the oral market is growing week over week. "We're confident long term" about Lilly's place in the pill segment, Ricks said, citing planned launches in additional international markets in the coming months.
Foundayo (搜索) entered the U.S. market in April. Lilly said in August that the pill recorded $98 million in sales for the second quarter, its first three-month period on the market. Medicare's new coverage of obesity (搜索) drugs, which began in July, is expected to broaden access to the pill and to Lilly's weight loss drug Zepbound.
Beyond GLP-1s: Oligonucleotides and Advanced Therapeutics
In addition to Foundayo (搜索), the Houston site will focus on advanced therapeutics that Lilly said were not within reach even a few years ago, including oligonucleotides, a class of medicines that can work at the genetic level to switch off the production of specific proteins to treat disease closer to its source. The plant will also manufacture API for Lilly's other small-molecule medicines across cardiometabolic health, oncology, immunology and neuroscience.
The site will use machine learning, AI, digitally integrated systems and advanced data analytics to drive right-first-time execution, with digital automation embedded throughout to streamline operations and support a reliable supply of medicines.
"This $6.5B investment will help change the game for tens of millions of people suffering from overweight, obesity (搜索) and its consequences like diabetes," Ricks said. "We will make and ship Lilly's latest products from Texas to people here at home and around the world."
Texas Governor Greg Abbott said the investment is the largest active pharmaceutical ingredient project in Texas history.
Workforce and Training Pipeline
Lilly expects the Houston facility to create more than 600 high-wage jobs, including engineers, scientists, operations personnel and lab technicians, along with 4,000 construction jobs as the site is built and brought online. The company is currently hiring in the Houston area.
Lilly is providing $12.5 million to support a relationship with San Jacinto College (搜索) to create pathways into advanced chemical synthesis manufacturing careers with no prior experience required. The collaboration will develop training pathways and expand equipment and facility capacity to produce technicians, operators, maintenance professionals and other manufacturing talent, with Lilly experts supporting curriculum development and instructor readiness. Separately, Lilly is donating $2.5 million to the San Jacinto College Foundation for scholarships for eligible students.
"When you invest in a place like Houston, you invest in its people first," said Edgardo Hernandez, Executive Vice President and President of Lilly Manufacturing Operations. "This facility will run on the talent of this community, powered by our relationship with San Jacinto College (搜索)."
Dr. Brenda Hellyer, Chancellor of San Jacinto College (搜索), said the relationship will expand industry-informed education and hands-on training pathways for students and incumbent workers.
Manufacturing Network Expansion
Lilly's ten announced U.S. sites since 2020 include five API facilities: Richmond, Virginia; Huntsville, Alabama; Houston, Texas; and two sites in Lebanon, Indiana, comprising Lebanon API and the Lilly Medicine Foundry, which will also focus on new production methods and making medicines for clinical trials. Four sites will make injectable medicines and manufacture devices: Research Triangle Park and Concord, North Carolina; Kenosha, Wisconsin; and Lehigh Valley, Pennsylvania. One site, Lebanon Advanced Therapies, will make genetic medicines.
Safety Profile
Foundayo (搜索) carries a boxed warning for thyroid tumors, including thyroid cancer. It is contraindicated in patients with a personal or family history of medullary thyroid carcinoma or Multiple Endocrine Neoplasia syndrome type 2, and in those who have had a serious allergic reaction to orforglipron or any of its ingredients. The label also warns of pancreatitis, severe stomach problems, dehydration leading to kidney problems, hypoglycemia, serious allergic reactions, vision changes in patients with type 2 diabetes (搜索), gallbladder problems, and food or liquid entering the lungs during procedures using anesthesia or deep sedation.
The most common side effects include nausea, constipation, diarrhea, vomiting, indigestion, abdominal pain, headache, swollen belly, fatigue, belching, heartburn, gas and hair loss. Foundayo (搜索) should not be used with other GLP-1 receptor (搜索) agonist medicines, and its safety and effectiveness in children have not been established. The drug is available as 0.8 mg, 2.5 mg, 5.5 mg, 9 mg, 14.5 mg and 17.2 mg oral tablets.
