The expanded agreement builds upon a strategic collaboration established in 2013 between the two companies, which enabled ZYNTEGLO's commercial approval in 2022. Under the renewed terms, Lonza will expand manufacturing capacity at its Houston facility, a dedicated cell and gene therapy site with nearly 10 years of combined clinical and commercial experience supporting the manufacture of this innovative treatment.
Growing Patient Demand Drives Expansion
Brian Riley, President and Chief Technology Officer at Genetix, highlighted the increasing patient need driving the partnership extension. "In the last year, patient demand for Genetix's therapies has grown consistently and rapidly," Riley stated. "Extending the agreement with Lonza and expanding capacity provides a strong foundation as we invest to meet the growing patient demand and broaden our long-term manufacturing strategy."
The collaboration also includes provisions to scale up manufacturing for additional Genetix therapies in the future, reflecting the company's broader pipeline of genetic treatments for severe rare diseases.
ZYNTEGLO's Clinical Impact
ZYNTEGLO (betibeglogene autotemcel) represents a significant advancement in treating transfusion-dependent beta-thalassemia (search). The therapy is a first-in-class, one-time ex-vivo lentiviral vector (LVV) gene addition therapy that adds functional copies of a modified beta-globin gene (search) (βA-T87Q-globin gene (search)) into a patient's own hematopoietic stem and progenitor cells (HSPCs).
This genetic modification results in the production of adult hemoglobin (HbAT87Q), enabling total hemoglobin levels to reach normal or near-normal levels. Clinical studies have demonstrated that ZYNTEGLO eliminates the need for regular red blood cell transfusions in eligible patients.
The FDA has recognized the therapy's significance by granting multiple designations, including orphan drug designation, fast track designation, regenerative medicine advanced therapy (RMAT) designation, and rare pediatric disease designation.
