If cleared by the FDA, the Phase 1/2a study will evaluate the safety, tolerability, dose optimization, and preliminary anti-tumor activity of Lm-LLO-TT (搜索) in patients with PDAC (搜索), one of the deadliest and most treatment-resistant solid tumors. The trial represents the first-in-human study for the company's lead therapeutic candidate.
Novel Approach to Treatment-Resistant Cancer
Lm-LLO-TT (搜索) is designed to stimulate tumor-directed memory immune responses in difficult-to-treat cancers. The platform utilizes engineered Listeria monocytogenes, a bacterium chosen for its ability to provoke robust cellular immune responses. This approach aims to mobilize the immune system against tumors that have historically resisted conventional treatments.

The therapeutic candidate was developed by Claudia Gravekamp, PhD, who has long explored the use of Listeria-based vectors in oncology. "Lm-LLO-TT (搜索) was created to mobilize the immune system in a way that could meaningfully change how we treat difficult solid tumors such as pancreatic cancer (搜索)," Gravekamp explained. "The promise of this platform lies in its potential to both engage memory immune mechanisms and target a tumor environment that has remained resistant to many existing approaches."
Addressing Critical Unmet Medical Need
Pancreatic cancer (搜索) remains one of oncology's most challenging diseases, with limited treatment options and poor patient outcomes. The dense, fibrotic and deeply immunosuppressive landscape of PDAC (搜索) has proven resistant to many therapeutic approaches, including checkpoint inhibitors (搜索) that have shown success in other cancer types such as melanoma (搜索) and lung cancers (搜索).
The therapeutic landscape for pancreatic cancer (搜索) demonstrates the urgent need for new approaches. Bradley highlighted the recent industry reaction to Revolution Medicines' daraxonrasib, which doubled median overall survival to 13.2 months in a late-stage trial. "It's being hailed as the best new medicine in the past 15 years," Bradley noted, emphasizing the limited progress in this indication over recent decades.
Platform Technology with Broader Potential
Beyond oncology applications, the Listeria-based platform may have implications for immune system restoration and cellular clearance. Bradley suggested the technology could potentially address senescent cell clearance using the immune memory system. "There's an opportunity to go beyond cancer to clearing other immune-suppressed cells, especially senescent cells, using your longer-term immune memory system," he explained.
The concept represents a shift from reactive cancer treatment to proactive immune maintenance. "I think this can become an annual treatment at some point, where you just clear out any bad cells before they become a serious illness," Bradley suggested.
Clinical Development Timeline
Matter Bio (搜索) expects to provide additional updates following completion of the FDA's review of the IND application. The company combines advanced tools for measuring DNA damage and genotoxicity with a pipeline of differentiated oncology programs aimed at addressing major unmet needs in cancer.
The IND submission represents the culmination of preclinical development work and positions the company to begin clinical evaluation of its lead oncology program.
