The validation study, titled "Simultaneous prenatal cfDNA screening of aneuploidy, recessive single-gene conditions, and fetomaternal blood compatibility," analyzed nearly 500 samples to assess the multiple components of the FirstGene screen. The comprehensive testing platform evaluates fetal aneuploidy (搜索), RhD compatibility, and recessive condition status in both the pregnant person and fetus, including screening for 19 prevalent and severe conditions.

Technical Innovation and Methodology
The FirstGene screen employs fetal-fraction amplification technology, similar to Myriad's Prequel test (搜索), to enhance accuracy. The platform incorporates a novel trajectory analysis algorithm specifically developed for the FirstGene screen to further improve genotype resolution. According to Dr. Muzzey, this approach provides "a thorough characterization of the ability to call fetal recessive genotypes—even technically challenging variants—with high accuracy."

Addressing Clinical Practice Gaps
Medical society guidelines recommend aneuploidy screening, recessive screening, and fetomaternal antigen testing as standard components of prenatal care. However, fewer than half of pregnancies in the United States receive screening for each of these recommended modalities individually, potentially driven by the logistical challenges of administering multiple separate tests.
The FirstGene screen aims to address this gap by streamlining prenatal genetic risk assessment through the combination of multiple testing modalities into a single assay performed from a single blood draw from the pregnant person.
Clinical Development and Commercial Path
FirstGene is currently being evaluated in the CONNECTOR study, a large, multi-site, prospective clinical study designed to evaluate test performance in real-world clinical practice. The study is specifically designed to generate evidence supporting clinical validity and clinical utility across the multiple components of the assay as FirstGene advances toward full commercial launch.
The test will be conducted in-house at Myriad's laboratories, requiring only a single blood draw from the pregnant person. According to Dr. Muzzey, "Publishing these results marks an important milestone on the path to broad commercialization."
