The multicenter, randomized, double-blind, placebo-controlled study involved 318 subjects across 9 clinical centers in France and was completed in 2024, with results published in July 2024 in Nicotine & Tobacco Research.
Clinical Trial Results Demonstrate Comparable Efficacy
The CESTO 2 study demonstrated efficacy comparable to that of other smoking cessation (搜索) treatments currently available. The primary endpoint was continuous abstinence for 4 weeks, with secondary endpoints including continuous abstinence for longer periods up to 12 months with a 12-month follow-up period.

Using positron emission tomography (PET) imaging in mice, researchers monitored changes in brain function associated with tobacco dependence development.
The study demonstrated NFL-101's unique and disruptive mechanism of action, showing the treatment's ability to reduce craving through restoration of normal brain activity in the area of the brain associated with craving. The results suggest communication between the immune system and the central nervous system, representing a mode of action different from current smoking cessation (搜索) drugs which directly target nicotinic receptors (搜索).
Simplified Administration Protocol
NFL-101 is administered via two subcutaneous injections, with the first injection coinciding with the target quit date and the second injection administered one week later. Potential for Combination Therapy
Based on earlier Phase 2a results from the PRECESTO study, NFL-101 could be offered to any smoker wishing to increase their chances of success, whether or not they are using other smoking cessation (搜索) treatments simultaneously. The PRECESTO study demonstrated that NFL-101 reduces smoking satisfaction, with the reduction being greater than that observed with Champix®/Chantix®.
Safety Profile Established
Preclinical studies and Phase 1 (CESTO) and Phase 2a (PRECESTO) clinical studies have endorsed the safety of NFL-101. The Phase 1 study conducted on 24 smokers confirmed the safety of the product with the absence of side effects, while demonstrating a specific action on the immune system.
The development of NFL-101 was initiated following the observation of the efficacy of a tobacco leaf allergy desensitization treatment in smoking cessation (搜索). These observations open up new perspectives not only in smoking cessation, but also in the treatment of related co-addictions involving tobacco, cannabis, and alcohol.
