Enhanced Trial Flexibility and Expanded Indications
The FDA agreement includes several key modifications that broaden the therapeutic potential of NKX019. Following final submission of amendments and IRB approvals, both Ntrust-1 and Ntrust-2 trials will enable outpatient administration and allow redosing options for patients who may benefit from additional treatment cycles. Additionally, rheumatoid arthritis (search) will be added as a new indication in Ntrust-2, expanding the study scope to address another autoimmune disease with significant unmet medical need.
The protocol amendments also eliminate geographic monitoring requirements, further reducing patient burden and operational complexity. These changes position NKX019 for broader accessibility while maintaining the rigorous safety standards required for cell therapy development.

Enrollment continues across both multi-center, open-label, dose-escalation trials, with patients now receiving 4 billion cells per dose as part of ongoing dose escalation. The current dosing regimen involves a three-dose cycle administered on days 0, 3, and 7 following lymphodepletion, delivering a total of 12 billion cells per treatment cycle.
The Ntrust trials are currently enrolling patients with lupus nephritis (search), primary membranous nephropathy (search), systemic sclerosis (search), idiopathic inflammatory myopathy (search), and ANCA-associated vasculitis (search).
