The transaction deepens an existing relationship between the two companies that began with a research collaboration and a single-asset license exercised in September 2024.
K-554 Leads the Acquired Portfolio
K-554 (搜索) is the most advanced of the three assets. Kallyope (搜索) describes the once-weekly peptide as targeting a previously undisclosed component of a neural circuit involved in feeding regulation, and has specifically distinguished its biology from both GLP-1 and amylin. A separate report describes K-554 as a non-incretin-based candidate currently in preclinical testing, with first-in-human Phase 1 trials slated for the second half of 2026.
The other two acquired programs are small molecules directed against separate novel targets and are in lead optimization.
The transaction gives Novo access to programs generated by Kallyope (搜索)'s neural circuits discovery platform, which is designed to identify signaling pathways linking peripheral organs with the brain and translate those circuits into drug targets. Kallyope has focused much of that work on the biological control of feeding and metabolism, providing Novo with mechanisms outside the incretin and amylin pathways that dominate much of the current obesity (搜索) pipeline.
A Different Discovery Axis for a Diversifying Pipeline
The assets broaden an obesity (搜索) pipeline that Novo has been deliberately diversifying beyond semaglutide. Alongside GLP-1-based therapies, the company is developing amylin programs including cagrilintide and zenagamtide (搜索), the CB1 inverse agonist monlunabant, and externally sourced multi-receptor agonists. The Kallyope (搜索) deal adds a discovery axis centered on neural control of appetite, with mechanisms that could potentially be developed alone or layered onto incretin therapy.
The pipeline expansion follows recent setbacks in Novo's internal development roster, including the discontinuation of a once-monthly GLP-1 candidate and the halting of a Phase 3 trial for ziltivekimab. A collaboration with Ascendis Pharma also came to an end, returning exclusive rights to TransCon metabolic programmes to the partner. Novo has also struck a licensing and development pact with Orbis Medicines (搜索) carrying milestone payments of as much as USD 1.4 billion for orally administered cardiometabolic macrocycles, and Petersen has signalled that several additional candidates are expected to enter clinical development over the course of the current year.
Kallyope's Platform and Parallel Pipeline
Kallyope (搜索) was founded in 2015 by Columbia University scientists Charles Zuker, Tom Maniatis, and Nobel laureate Richard Axel to translate emerging research on gut-brain signaling and neural circuits into therapeutics. The company built a discovery platform combining technologies including single-cell sequencing, computational biology, optogenetics, chemogenetics, and circuit mapping to identify druggable pathways linking peripheral organs with the brain.
The biotech has attracted substantial venture backing since launching with a USD 44 million Series A, going on to raise over USD 400 million over three rounds from investors including Lux Capital, Polaris Partners, The Column Group, and Mubadala. Kallyope (搜索) is continuing to advance its own pipeline alongside the Novo transaction: lead asset elismetrep (K-304), a first-in-class oral TRPM8 channel blocker for acute migraine, has progressed into Phase III after positive Phase IIb results. The combination of a late-stage internally developed migraine program and multiple obesity (搜索) assets now acquired by Novo provides clinical validation for Kallyope's neural-circuits discovery platform.
Regulatory Momentum Alongside the Deal
Novo's pipeline activity has arrived alongside regulatory progress in Europe. The European Medicines Agency's Committee for Medicinal Products for Human Use backed a marketing authorisation for FREHEMGO (denecimig) in haemophilia A (搜索) for both adults and children. The therapy is the first FVIIIa mimetic to offer prophylaxis in a ready-to-use pen at intervals of once weekly, every two weeks, or once monthly. Supporting data appeared the same day in the Journal of Thrombosis and Haemostasis, where results from the Phase 3b FRONTIER5 study showed favourable tolerability following a direct switch from emicizumab to denecimig, along with sustained thrombin generation within the normal range. Novo plans to launch FREHEMGO in the first European countries in the fourth quarter of 2026, widening the rollout across the EU from early 2027.
A second positive opinion followed, with the CHMP recommending approval of the growth hormone Sogroya (somapacitan) for weekly treatment of children with idiopathic short stature (搜索). If the European Commission follows the recommendation, with a decision anticipated before year-end, Sogroya would become the first and only therapy specifically authorised for this indication in the EU.
Novo has also unveiled a collaboration with Anthropic (搜索) aimed at tackling drug-discovery challenges using artificial intelligence, streamlining scientific workflows and speeding up the development of new medicines, and rolled out a unified brand identity under the name "Novo."
