Opterion's OPT101 Clears Phase 1 in Peritoneal Dialysis, Advancing to Phase 2a GLOW Trial

The Swiss clinical-stage biotechnology company said the study achieved its objectives across all major endpoints, including safety, tolerability, pharmacokinetics and functional activity in patients with moderate to severe chronic kidney disease (搜索), supporting advancement of the program to Phase 2a.
SPARC Design and Endpoints
SPARC evaluated different concentrations of OPT101 in four-hour and eight-hour dwells in patients with moderate to severe chronic kidney disease (搜索). The study reported no serious adverse events, no treatment discontinuations due to adverse events, no dose interruptions, no concentration-limiting toxicities and no interruptions to dose escalation.
OPT101 demonstrated activity across all evaluated concentrations. The highest concentration produced the greatest mean fluid removal in both short and long dwells, accompanied by sodium removal. Fluid and sodium removal across both dwell settings underpins the company's positioning of OPT101 as a differentiated non-glucose-based approach to peritoneal dialysis.

Investigator Perspective on the Unmet Need
Aivars Pētersons, Principal Investigator of the SPARC study, described the trial as a positive first clinical experience with OPT101 and said patients responded well. "Peritoneal dialysis offers patients greater independence, but current solutions still involve important limitations," Pētersons said. "A flexible, non-glucose-based option for both short and long dwells could address a clear unmet need and be a welcome innovation."
Peritoneal dialysis allows people with kidney failure (搜索) to receive life-sustaining treatment at home, but patients still rely heavily on glucose-based solutions that can contribute to metabolic complications, weight gain and damage to the peritoneal membrane over time, according to the company. Existing non-glucose alternatives cannot cover both short and long dwells, which limits treatment choice and adds complexity to an already demanding daily routine. Opterion states that no currently available solution offers the same intended combination of non-glucose-based treatment, concentration flexibility and use across both dwell settings.
Phase 2a GLOW Trial Planned
The Phase 2a GLOW trial is expected to evaluate repeated administration of OPT101, dose selection, broader dwell replacement and the agent's functional and metabolic differentiation in chronic kidney disease (搜索) patients.

Peritoneal dialysis accounts for only around 12% of dialysis patients globally despite offering clinical and lifestyle advantages, according to Opterion.
