Plant Virus Nanoparticles Show Broad Protection Against Metastatic Cancers in Preclinical Studies

Researchers at the University of California San Diego have developed a novel cancer immunotherapy using nanoparticles derived from a plant virus that shows broad protective effects against metastatic cancers in preclinical studies. The experimental treatment, composed of cowpea mosaic virus (search) nanoparticles, demonstrated remarkable success in improving survival rates and suppressing metastatic tumor growth across multiple cancer types in mouse models.
The findings, published in Advanced Science, represent a significant advancement in the work led by Nicole Steinmetz, professor of nanoengineering and director of the Center for Nano-ImmunoEngineering (search) at UC San Diego (search). The research builds upon previous studies where cowpea mosaic virus (search) nanoparticles were injected directly into tumors to stimulate immune responses.
Systemic Administration Proves Effective
The breakthrough finding of the current study is that the plant virus nanoparticles do not require direct tumor injection to be effective. Systemic administration of the nanoparticles improved survival rates and inhibited metastasis across various cancer types, marking a significant step toward clinical applicability.
"Here, we do not treat established tumors or metastatic disease—we prevent them from forming. We are providing a systemic treatment to wake up the body's immune system to eliminate the disease before metastases even form and settle," said Steinmetz.
The treatment leverages the immune system's natural response to foreign pathogens. Although the cowpea mosaic virus (search) is non-infectious in mammals, immune cells recognize it as foreign, triggering a robust immune reaction against existing tumors and providing protection against future tumor formation.
Manufacturing and Testing Approach
To produce the nanoparticles, researchers grew black-eyed pea plants in the laboratory and infected them with cowpea mosaic virus (search). Millions of copies of the virus were harvested in the form of ball-shaped nanoparticles, requiring no further modification before experimental use. Steinmetz described these as "Nature's powerful nanoparticles, as produced in black-eyed pea plants."
The research team tested the treatment's efficacy in mouse models of colon, ovarian, melanoma (search), and breast cancers. Mice injected with cowpea mosaic virus (search) nanoparticles and then challenged with metastatic tumors one week later exhibited improved survival rates and reduced tumor growth compared to untreated controls. The protective effect persisted when mice were challenged with new tumors a month after initial treatment.
Post-Surgical Applications Show Promise
The researchers found particularly encouraging results in post-surgical applications. In experiments where nanoparticles were administered after surgical tumor removal, treated mice showed improved survival rates and decreased tumor regrowth compared to controls.
"Even if you perform surgery to remove the tumors, no surgery is perfect and there is outgrowth of metastasis if no additional treatment is provided," explained Steinmetz. "Here, we use our plant virus nanoparticles after surgery to boost the immune system to reject any residual disease and prevent circulating tumor cells from metastatic seeding. We found that it works really, really well!"
Clinical Translation Plans
The research team is now preparing for clinical trials as the next phase of development. Planned activities include conducting safety studies and exploring the treatment's efficacy in pet animals with cancer. Future research will also focus on understanding the underlying mechanisms of the immune-boosting properties of cowpea mosaic virus (search) nanoparticles.
