Pneumagen's Neumifil Shows Promising Results in Phase 2 Influenza Prevention Trial
核心洞察
Pneumagen (搜索)'s intranasal antiviral drug Neumifil demonstrated clinically significant reductions in both incidence and severity of symptomatic influenza (搜索) infections in a Phase 2 controlled human infection model study.
The broad-spectrum antiviral, developed using proprietary GlycoTarge technology, works by directly binding to viral pathogens (搜索) and preventing their entry into host cells.
Based on these positive results presented at the American Thoracic Society meeting, Pneumagen (搜索) plans to advance Neumifil into further clinical studies, including evaluation for reducing COPD (搜索) exacerbations in high-risk patients.
Pneumagen (搜索), a clinical-stage biotech company based in St Andrews, Scotland, has reported positive Phase 2 results for its broad-spectrum antiviral drug Neumifil (HEX17), demonstrating clinically significant reductions in symptomatic influenza (搜索) infections. The findings were presented at the American Thoracic Society meeting in San Diego on May 21, 2024.
Study Design and Results
The Phase 2 proof-of-concept study utilized a controlled human infection model (CHIM) where healthy volunteers were challenged with influenza (搜索) virus following administration of Neumifil. The randomized, double-blind, placebo-controlled study was conducted at a single UK center and evaluated two dosing regimens: a single dose administered three days before infection, or three daily doses on the three days leading up to infection.
Results showed that Neumifil led to notable reductions in both the incidence and severity of symptomatic influenza (搜索) infections compared to placebo. The intranasal spray also considerably decreased viral load in treated participants.
"Viral respiratory infection is of enormous global concern, particularly in patients with underlying pulmonary disease, in whom viral infections can induce exacerbations," said Dr. Geoff Kitson, Pneumagen (搜索)'s Chief Medical Officer and presenting author. "The significant reductions in both symptoms and viral load demonstrate the potential of this new antiviral drug not only to alleviate patients suffering but also to curtail the spread of virus within communities."
Novel Mechanism of Action
Neumifil is a novel, engineered, multivalent carbohydrate binding module (mCBM) developed using Pneumagen (搜索)'s proprietary GlycoTarge technology. The drug acts by directly binding to viral pathogens (搜索) and preventing their entry into host cells, offering broad-spectrum antiviral activity for the prophylaxis and treatment of viral respiratory tract infections (搜索).
The safety and tolerability profile of Neumifil was consistent with previous human studies, supporting its potential for broader clinical application.
Future Development Plans
Based on the promising proof-of-concept results, Pneumagen (搜索) plans to advance Neumifil into further clinical studies. "These trials are expected to include the evaluation of Neumifil's ability to reduce the incidence of exacerbations in patients with Chronic Obstructive Pulmonary Disease (搜索) (COPD (搜索)), which represents a significant at-risk population," said Douglas Thomson, Pneumagen's CEO.
The company's strategy leverages these positive Phase 2 results alongside preclinical data to expedite Neumifil's development as a versatile antiviral treatment against various viral infections. Future investigations will aim to confirm the drug's potential as a key therapeutic option for prevention and treatment of respiratory tract infections, particularly in high-risk patient populations.
Company Background
Pneumagen (搜索) was founded in 2016 as a spin-out from the University of St Andrews and benefits from extensive scientific expertise in viral infections, particularly in glycobiology. The company is developing a pipeline of antiviral therapies targeting diverse infectious diseases using the GlycoTarge platform technology.
