The partnership represents a novel approach to addressing one of genetic medicine's most challenging problems: designing personalized genomic interventions for rare disease patients.
AI-Powered Gene Editing Approach
The collaboration will leverage Profluent's frontier AI models to engineer potent and specific base editors targeting recurrent "hot-spot" mutations in the MeCP2 (search) gene that cause Rett syndrome (search). These novel editors will be designed to be compact enough to fit inside a single adeno-associated virus (AAV) vector, which is critical for efficient delivery and gene correction in the central nervous system.
"Our AI is built to navigate a vast design space and find solutions that are out of reach with manual protein engineering methods, resulting in personalized mutation-specific genomic medicines that are an ideal match for each Rett patient," said Peter Cameron, Profluent's Senior Vice President of Gene Editing and Translation.
The initial focus will be on the T158M (search) mutation, a commonly observed variant in Rett patients. The approach aims to permanently correct inherited mutations rather than providing temporary therapeutic relief.
Addressing Unmet Medical Need
Rett syndrome (search) is caused by random mutations in the MECP2 (search) gene on the X chromosome and typically becomes apparent between 12 to 18 months of age. The disorder deprives toddlers of speech, hand use, and normal movement, often including the ability to walk. As the condition progresses, patients experience anxiety, seizures (search), tremors, breathing difficulties, and severe gastrointestinal issues, while their cognitive abilities are believed to remain largely intact.

The partnership addresses a fundamental challenge in rare disease therapeutics: the tension between addressing rare diseases and making the economics viable. RSRT (search) serves as the largest funder of Rett research worldwide, having raised almost $110 million since its founding. The organization is the only non-profit exclusively focused on advancing genetic medicines for Rett syndrome (search). Multiple clinical trials in gene therapy for Rett syndrome are currently underway, all made possible through RSRT-funded research.
In 2024, RSRT (search) launched a $40 million initiative called "Roadmap to Cures," with the ambitious goal of bringing three genetic medicines beyond current gene therapies to clinical trials by 2028. This collaboration with Profluent represents a key component of this strategic initiative.
