Python-Derived Metabolite Shows Promise as Novel Weight Loss Drug with Reduced Side Effects

Researchers at the University of Colorado Boulder have identified a promising new approach to weight loss therapy by studying the extraordinary metabolic capabilities of pythons. The team discovered that a metabolite called para-tyramine-O-sulfate (search) (pTOS (search)), which increases dramatically in python blood after feeding, acts as an effective appetite suppressant in mice without the common side effects associated with current weight loss medications.
The research, published in Nature Metabolism, emerged from a collaboration between CU Boulder, Stanford University, and Baylor University scientists who sought to understand how pythons can survive extreme feeding cycles—consuming meals equivalent to their entire body weight and then fasting for months or even years without losing muscle mass or experiencing metabolic damage.
Metabolic Extremes Inspire Drug Discovery
"This is a perfect example of nature-inspired biology," said Leslie Leinwand, distinguished professor of Molecular, Cellular and Developmental Biology at CU Boulder and senior author of the study. "You look at extraordinary animals that can do things that you and I and other mammals can't do, and you try to harness that for therapeutic interventions."
Leinwand has been studying pythons in her laboratory for two decades, fascinated by their ability to increase their metabolism by 4,000 times and expand their hearts by 25% in the hours following a meal to aid digestion. After consuming a massive meal, pythons can return to their baseline metabolic state with no apparent damage to their cardiovascular system or loss of muscle mass during subsequent fasting periods.
Breakthrough Metabolite Discovery
The research team collected blood samples from ball pythons and Burmese pythons immediately after feeding for 28 days. Their analysis revealed that 208 metabolites were dramatically elevated following the pythons' meals, but one molecule stood out: para-tyramine-O-sulfate (search) showed a remarkable 1,000-fold increase.
When researchers administered high doses of pTOS (search) to both obese and lean mice, the compound affected the appetite center of the brain, specifically targeting the hypothalamus (search). The treated mice ate less and lost weight without experiencing gastrointestinal problems, muscle loss, or declines in energy—side effects commonly associated with current GLP-1 weight loss drugs like Ozempic and Wegovy.
"We've basically discovered an appetite suppressant that works in mice without some of the side-effects that GLP-1 drugs have," Leinwand explained.
Commercial Development and Future Applications
Building on these promising results, Leinwand has partnered with Stanford University professor Jonathan Long, who specializes in research on blood chemicals related to energy consumption and expenditure, and researchers at Baylor University who conducted the mouse studies. Together, they have formed Arkana Therapeutics (search) to develop the discoveries into commercially available drugs.
"If we truly want to understand metabolism, we need to go beyond looking at mice and people and look at the greatest metabolic extremes nature has to offer," Long said.
The company received a $127,500 grant last year to fund its work, and Leinwand's previous success in drug development—having co-founded two companies that sold for billions of dollars to pharmaceutical companies—provides valuable industry connections and venture capital access.
Broader Therapeutic Potential
Beyond weight loss applications, the researchers believe pTOS (search) and other python metabolites could address additional health conditions. The team plans to investigate whether the metabolite could treat age-related muscle loss, or sarcopenia (search), and intends to study other metabolites that increase after pythons eat.
"We're not stopping with just this one metabolite," Leinwand stated. "There's a lot more to be learned."
The research represents a significant advancement in nature-inspired drug discovery, demonstrating how studying extreme biological adaptations can lead to novel therapeutic approaches. As the team works to develop synthetic versions of these compounds, the python-derived metabolites could offer new hope for patients seeking effective weight loss treatments with improved safety profiles.
